PurposeWe use a new segmental mouse pancreas transplant model to detect the protection effect and the mechanism of cisplatin in a pancreas Ischemia/Reperfusion Injury model of mouse.MethodsWe chose 24 male Balb/c mice aged 6 to 8 weeks, randomly assigned to 2 groups, group I and group II. Each group got 12 mice, and every two mice were made donor and recipient pair. All donors in group I were injected with 0.5ml NS from the femoral vein, 12hs before the surgery, while donors in group II were given the same volume cisplatin, 1mg/kg body weight, from the femoral vein,12hs before the surgery.Then all transplantations were performed by same surgeons.All grafts were harvested 6hs after the transplantation, all samples were performed with H&E staining, MPO staining, ICAM-1 staining and TUNEL staining, and Western Blot was performed to detect the expression of HMGB-1 and IL-6 in all grafts.All datas were compared using T-Test, and the Western Blot results were analyzed using IPP software.ResultsAll grafts in group I showed severer edema and inflammatory infliction than that in group II, paralleled with more cell apoptosis. Western Blot showed both HMGB-1 and Il-6 in grafts of group I expressed more than those in group II.ConclusionCisplatin plays a protection role in mouse pancreas IRI by reducing the graft edema and Inflammatory cytokines releasing, HMGB-1 and Il-6 in that study/...
