Objective:Tanreqing injection is a Chinese medicine made up of scutellaria, bear gall powder, cornu caprae hircus powder, honeysuckle and fructus. According to the literatures, the drug shows ability against various virus and bacteria and always treated with several infections combined with other antibiotics, including anti-infection effects against infections followed with leukemia. However the effect of Tanreqing injection on the leukemia cells is still unknown. The present study, the inhibition effect of Tanreqing on K562, Molt 4 and Dami cell line was investigated and the influence on cell cycle and apoptosis of Molt4 cell line was also determined. Furthermore, the mechanism of apoptosis of leukemic mediated by Tanreqing injection was analyzed.Materials and methods:Tanreqing injection was folded dilution into 9 groups with various vaules, i.e.,1:16,1:32,1:64,1:128,1:256 and 1:512. The K562, Molt 4 and Dami cells were treated with above amounts of Tanreqing injection. The morphology of cells was observed using microscope. Cell proliferation was employed by CCK-8 assay and growth curve was acquired for calculating inhibition ratio and 50% inhibiting vaule (IC50). Molt4 cells treated using Tanreqing injections was tested with different levels reagent such as 1:32, 1:64,1:128,1:256,1:512 vaules at 24h,48h and 72h, respectively. Cell cycles and apoptosis were analyzed using FCM; Furthermore, Molt 4 cells treated with Tanreqing injections of 1:32,1:64,1:128,1:256 and 1:512 vaules at 72 h, And the apoptosis genes caspase 3 and bcl-2 were analyzed by qPCR method, Moreover, he possible mechanism.Results:1. K562 cells, Molt 4 cells and Dami cells can be inhibited by Tanreqing injection depending on vaule.2. Compared with control, cell cycle of Molt4 cells can be inhibited by Tanreqing injection in 1:32 vaule (P<0.01).3. Compared to control group, Molt 4 cells can be induced apoptosis by Tanreqing injection in 1:32 vaule (P<0.01).4. Comparing the control, genes that caspase 3, bcl-2 andβ-actin decreased after Molt 4 cells treated with Tanreqing injection 48 h in 1:32 values.Conclusion:1. Tanreqing injection exhibits inhibited effects against leukemia depend on drug amounts.2. Compared with control group, cell cycle of Molt 4 can be significantly inhibited by Tanreqing injection in 1:32 vaule.3. Apoptosis of Molt 4 cells can be induced by Tanreqing injection in 1:32 vaule which there is no relationship with the bcl-2 -aspase 3 pathway.4. Molt 4 cells backbones are destroyed by Tanreqing injection in 1:32 vaule and the cells segmentations were inhibited.
