The restoration of animal tissues and organs after injury is an important function in vital movement. The stem cell which is in the adult tissues and organs is considered to the main force. The cell usually lose the ability of resegmentation in the reason of high degree cell differentiation when splits apart in the embryonic period. Organism keeps some undifferentiation initial cells which are called Adult stem cells ASCs to compensate the fault. These stem cells would produce differentiation cells by the segmentation according to the development paths when are needed. Therefore, they are the reserve of the differentiation cells. Stem cell has potential abilities and self-renewal but at a slow multiplication. The mitotic cycle of stem cells in the adult organism is not indefinite.Stem cell has regulatory factors which react the external signals to regulate the proliferation and the differentiation. In other words, it has the character of endogenous control. The surrounding tissues and cell epimatrix would influence the cell differentiation. In addition to this, the adult stem cell has a strong plasticity when transplanted into the receptor. Stem cell will be influenced by the surrounding ongoing differentiation cells and regulate the reactions to the differentiation signals to react the regulate signals in the new microenvironment when it comes into the new microenvironment. Other scholars believe that the potential differentiation abilities are different form the pluripotency of the embryo stem cell. The plripotency usually be definite in the identical embryonic layer or several cell types in the identical system. Therefore, it is called the pluripotency in the single embryonic layer in contrast to the stem cell has the pluripotency in the three embryonic layers.Whether this fact would withstand the text of time, the landscape orientation differentiation of stem cell merely definite the in vitro now, it is not found any reports about the step across differentiations when in normal or injury-repair in vivo. But, some domestic scholars suggest the new viewpoint that signal tissue vectoring stem cell could not preserve the quick renewal. There must be more stable cell resources to preserve the quantity and quality of the vectoring stem cells.The stem cell of the connective tissue in the mesoderm resource differentiates to endoblast and ectoderm to the vectoring stem cell. But this process is not be observed easily, so the scholars suggest that one of the function of connective tissue is store stem cells and these cells differentiate to different function cells regularly to repair and preserve the healthy and stable of the whole organism.Cell proliferation is one of the basic characteristics of cells, proliferation includes the cell grow, DNA copy and segmentation, it achieves through the cell cycle.BrdU is mixed with the DNA molecules which are copied during the S phrase in the cell cycle. When the immune tissue samples are chemical staining, it combines with the BrdU antibody. Thus, this could detect the cell that copying DNA vigorously which called proliferation cell. Methenamine silver basilar membrane stained method could illustratethe position which around by dark brown basilar membrane to observe whether cell from dermal connective tissue perceive basilar membrane.Purpose of the experiment: investigate the rationality of the new viewpoint about the stem cell of the connective tissue in the mesoderm resource differentiates to endoblast and ectoderm to the vectoring stem cell by studying rats.Empirical approach: several Healthy male Wistar rats are divided into two groups, one is the injury epithelial tissue, and the other is normal group by contrast. The injury epithelial tissue includes gastric mucous injury group, skin scratching group and skin incision group. Three randomly selected rats for test preparation. The two groups are HE strained, Immunohistochemistry and Methenamine silver stained the basement membrane.Experimental results: (1) HE staining showed that normal gastric epithelial is complete, the absorb cell straining is vivid and bright. Gastric parietal cells and chief cell in the proper player are normal, gastric wall layers are demarcated. Gastric mucous injury in the impairment is space-occupying lesion and little inflammatory exudation and hyperaemia. Skin incision is showed after HE strained. Surrounding tissue incision are hyperaemia and edema, inflammatory cell is soakage. Dermal connective tissue fibers in skin scratching tissue is relax, severely congestive, no other abnormalities. (2). Gastric mucosa after BrdU immune tissue chemical strained, fundic gland in normal gastric mucous injury could find little BrdU immune reactions in the nucleolus. There are lots of BrdU immune masculine cells in the fundic gland of the stress ulcer. There is no BrdU masculine cell in the proper layers between the fundic glands. Skin BrdU immunohistochemistry is good, and there is no BrdU immunoreactive cell in the skin incision surrounding tissue, neither in the skin scraping group. (3). Methenamine silver staining the basement membrane successes. There is clear and complete dark brown basement membrane round the normal stomach peripheral in the longitudinal and transverse section of normal stomach. The fundic gland around the gastric mucous injury position could be seen the complete basement membrane circumvolution. No damage methenamine silver staining of skin basement membrane successes, basilar membrane in the junction of epidermis and dermis is vivid and complete. There are tissue defects in the skin incision and inflammatory cell infiltration, but no basilar membrane. Local cell layers around the skin incision increase. Basement membrane may not complete, scraping the skin after skin deep also has similar Morphological features.Conclusion: 1. proliferative cells are more than the normal control group in gastric mucous injury cells of the Stress ulcer mucosa, fundic gland basement membrane is clear and complete, and there is no penetration of the basement membrane lamina propria connective tissue cells.2. There is no proliferative cell 32 in skin incision and scraping the skin tissue in epidermis and dermis, no dermal connective tissue cells also penetrate into the epidermal basement membrane. The new viewpoint about the stem cell of the connective tissue in the mesoderm resource differentiates to endoblast and ectoderm to the vectoring stem cell subject to further discussion.
