Atrial fibrillation is common as a kind of chronic arrhythmia.It usually happenes to the patientes who has organic heart diseases, a few clear out patientes also can exist. The incidence of 1,000 women, men for weaker 21.5 per 1,000 and increase with age, its incidence increased significantly. Along with the heart structure change, heart failure, malignant arrhythmia, blood clots, heart, and complications of thrombosis, emerge in endlessly, AF is a serious disease to the human life quality. So far, there have been many scholars to clear the mechanism and the prevention of atrial fibrillation, a lot of basic research and clinical observation, proposed many hypothesis and treatment.They have made certain progress and success, but many problems of the mechanism, it is still not fully understood.The sleeve muscle might for heterotopic excitement of atrial fibrillation and atrium electrophysiology reconstruction and structure for the reconstruction of atrial fibrillation may occur, and maintain the important condition. In the reconstruction, atrial ionic channel, protein and atrium of energy metabolism change is a research hotspot. And proteomics has the accuracy and operability, quickness and universality in the protein study now, leading and pillar status. Application of proteomics research methods of atrial fibrillation in patients with atrial muscle protein expression, modification, etc, to reveal the occurrence of af, mechanism, and has the vital significance to further help atrial fibrillation prevention, treatment and prognosis of improvement of the development of the research, etc.Objective:to chronic atrial fibrillation and sinus rhythm RenXinFang muscle tissue proteomic technologies applied to analysis, comparison, in protein level af mechanism, and further promote the prevention and treatment of atrial fibrillation.Methods:Randomly selected from November 2009 to March 2010 in our hospital undergoing open heart surgery operation of chronic atrial fibrillation (4 patients) and sinus rhythm (four cases) and atrial muscle as the experimental samples, the atrialin liquid nitrogen into powder form, the application of lysis buffer cracking, and with the Clean-up Kit Kit to the crude protein purification. Spectrophotometer to measure and calculate the protein content of samples.Will extract samples of heart muscle proteins fluorescently labeled two-dimensional electrophoresis (isoelectric focusing in the first row will be different proteins according to isoelectric point of separation, re-SDS-polyacrylamide gel electrophoresis, separated according to molecular weight).Electrophoresis finished scanning.Then scan the images obtained were compared and analyzed.Finally, comparison of protein database obtained by the differentially expressed proteins.Results:Spectra of the control group by image analysis of myocardial average detected (1,319±13) protein spots for the experimental group (1,332±16) protein spots.The statistical analysis were 12 significantly different protein spots.Conclusion:When atrial fibrillation compared with sinus rhythm, the differentially expressed proteins, and these proteins may be related to the occurrence of atrial fibrillation and sustained.
