Object:.To study the inhibition effect for apoptosis and invasion of transfection of antisense oligodeoxynucleotide (ASODN) targeting Survivin gene on hilar cholangiocarcinoma cell line FRH-0201.To investigate the level of MMP-2 and TIMP-2, and investigate the invasion change of the cell.Methods:Three groups were divided during the experiment,the ASODN group,the SODN group, the blank control group. The ASODN was transfected into Cholangiocarcinoma cell line FRH-0201 by LipofectamineTM2000.Detecting the level of MMP-2 and TIMP-2 in the supernatant by enzyme linked immunosorbent assay (ELIS A).The effect for invasion of transfection of ASODN targeting Survivin gene on cholangiocarcinoma cell was observed with Transwell cell culture chamber.Results:1.Morphological apoptotic changes were observed. The cell line's proliferation was inhibited in the ASODN group.2. The expression of MMP-2 in ASODN group was lower than that in the SODN and blank group (61.32±2.21vs94.15±3.32; 61.32±2.21vs100.45±5.86,P<0.05), the level of the TIMP-2 in the ASODN group was higher than that in the other groups (95.01±1.36vs58.21±4.42; 95.01±1.36vs60.92±5.47,P<0.05).3. The number through the membrane of the cell culture chamber was decreased in ASODN than that in the other two groups (21.33±4.16vs46.33±3.51; 21.33±4.16vs43.67±2.08.P<0.05).Conclusions:The ASODN targeting Survivin Gene can significantly inhibit the cell proliferation and invasive ability,and induce apoptosis of the FRH-0201.
