Objective:We study the changes of CD34 and BCL-2 in the right atrial tissues during cardiopulmonary bypass surgery in early myocardial ischemia-re perfusion. The study reveals the correlation of the vascular endothelial cell injury and the anti-apoptotic capacity of myocardial cells.The result can be a reference for the myocardial protection during card- iopulmonary bypass and the myocardial stem cell transplantation.Methods:We harvested the right atrial tissues of patients for cardiopulmonary bypass surgery in the No.1 hospital of Longyan City, affiliated to Fujian Medical University in 2009.02~2009 .10. The HE and the immunohist- ochemical staining were use to find CD34 and BCL-2 staining positive cells .Then, we used statistical analysis to deal with the positive cells and observed the changes with time. All the data were analysized by SPSS 13.0 statistical software. The following statistical methods: mean and standard deviation, X2 tests, Wilcoxon rank sum test, multivate analysis of variance, Pearson correlation analysis were used.Results:1.The study showed that the CD34 was decreased with time,and BCL-2 was in upward trend in ischemia stage and decreased in the early stage of reperfusion, the difference was statistically significant ( P <0.05). 2.The correlation analysis showed that the CD34 and BCL-2 was negatively correlated (P <0.05) in ischemia stage;and was positively correlated (P <0.05) in the early reperfusion stage, and the longer in ischemia time the more significant in the expression. Conclusions :The decreased CD34 expression in the ischemia stage means the injury of vascular endothelial cell during cardiopulmonary bypass surgery, while the increased BCL-2 expression in myocardial cells shows the enhanced ability of anti-apoptosis. But in the early reperfusion stage decreased CD34 and BCL-2 reveal the increasing injury of the endothelial cell and the reducing of myocardial anti-apoptosis ability. Therefore reducing endothelial cell injury in ischemia stage did not enhance the myocardial anti-apoptosis ability, but improving endothelial cells protection could enhance the ability of myocardial cells anti-apoptosis in the early stage of reperfusion.We conclude that less injury of vascular endothelial and more protection of endothelial cells could enhance the ability of myocardial self-protection in early stage of reperfusion during cardiopulmonary bypass surgery.
