Objective Evidence from family and epidemiology suggest that genetic factors are likely to play an essential role in the developing of ischemic stroke. It is a complicated disease caused by several genes and enviroment factor. it is an useful strategy to determine potential candidate genes of ischemic stroke for the prevention and individualized treatment. The purposes of this experiment is to evaluate the distribution of MCP-1 gene-2518G/A polymorphism in Han population of Shanxi province and the ethnic differences of this polymorphism. To study the association between monocyte chemoattractant protein-1 gene polymorphism and ischemic stroke.Methods To select 224 cases of ischemic stroke patients and 112 cases of healthy people as control group in Han population of Shanxi province. The MCP-1-2518-site single-nucleotide polymorphism was detected by using phenol-chloroform method to extract the genomic DNA, polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) and direct gene sequencing. we calculated the genotype and the allele frequency of MCP-1 in the different ethnic or the region of the world and compared the genotype and allele frequencies among groups to analyze the association with ischemic stroke and clinical variables. Compared clinical variables between groups using t-test and chi-square test and frequencies among groups by chi-square test. All data were used to process by SPSS11.3.Results1 The genotype distribution frequency of MCP-1 gene in Han population of Shanxi province is GG 37.5%, GA 50.0%, AA 12.5% and allele frequency is G 62.5%, A 37.5%.2 There is significant difference comparing MCP-1-2518 G/A genotype, allele frequency in the healthy Han population of Shanxi province to the people of Hunan province, but no significant difference to the people of southern Jiangsu and Harbin province. There is significant difference when compared with Slovakia, Tunisian, Spain, Turkey population and no significant difference compared with the South Korean population.3 MCP-1 gene-2518G/A genotype frequency in Han population of Shanxi province whom had ischemic stroke is GG 41.1%, GA 44.6%, AA 14.3%; G allele frequency is 63.4%, A allele frequency is 36.6%. And compared to the control group, there is no significant difference in the genotype and allele frequency.4 Compared with the AA genotype, MCP-1 gene GA+GG genotype had higher risk to have ischemic stroke after adjustment for sex, age and other well-established risk factors by binary logistic regression analysis, but there is no significant difference.5 There is no significant difference in the genotype and allele frequency between hypertension and non-hypertension, diabetes mellitus and non-diabetes, high LDL hyperlipidemia and non-high LDL hyperlipidemia in patients with ischemic stroke.Conclusions1 There is MCP-1 gene-2518G/A single nucleotide polymorphism in Chinese Han population of Shanxi province.2 MCP-1 gene-2518G/A single nucleotide polymorphism has racial, regional difference.3 There is no correlation between MCP-1 gene-2518G/A polymorphism and ischemic stroke in Han population of Shanxi province.4 There is no correlation between MCP-1 gene-2518G/A polymorphism and hypertension, diabetes mellitus, high LDL hyperlipidemia in Han population of Shanxi province.
