| Objective To investigate levels of CTLA-4 on peripheral blood Treg cell in patients with rheumatoid arthritis(RA) and in a control group of subjects, and analyse its relationship with IL-10,TGF-β1 and DAS28. The characteristics of CTLA-4 in RA were discussed.Method 30 patients with RA as defined by ACR criteria were investigated as study group.The control group consisted of 30 healthy volunteers.The disease activity was measured by the DAS28. Levels of CTLA-4 on peripheral blood Treg cell assay was performed using FLOW CYTOMETRY.Serum IL-10, TGF-β1 assays were performed using ELISA. Three groups were analyzed using ANOVA,two groups were analyzed using t test. Pearman's correlation was used in related analysis.Results①CTLA-4,IL-10,TGF-β1 values of the patients group were significantly different compared with the control group,P<0.01;②The levels of CTLA-4,IL-10,TGF-β1 among active, non-active group and healthy subjects were significantly different,P<0.01;③Related analysis: There was a significant correlation between CTLA-4 and IL-10, TGF-β1 in RA group. There was a negative correlation between CTLA-4 and DAS28 in RA group.Conclusion Lack of CTLA-4 on Treg cell may have an important role in the pathogenesis of rheumatoid arthritis,and can reflect disease activity. |
