Objective:To investigate the effect of ursodeoxycholic acid on liver regeneration and cyclin D1 mRNA and protein expression in rats.Methods: Male Wistar rats were randomly divided into two group:the control group in which the rats was fed standard diets in and the ursodeoxycholic acid (UDCA)group in which rats was given 0.1% UDCA diets. After 7 days on those diets,70% partial hepatectomy(PH) were performed. On 0,1,2 and 3 days after PH, the rat liver regeneration was examined by immunohistochemical method; the mRNA expression of cyclin D1 was examined by Quantitative RT-PCR and the expression of cyclin D1 protein was detected by western blotting.Results:On day 0, the PCNA hardly moved in all rats on day 0 after PH. On day 1, the PCNA labeling index reached a peak, and it was significantly higher in UDCA group than the control group(40.70±4.73 vs 33.24±5.59, P<0.01). On day 2, PCNA labeling index in UDCA group was still significantly higher than the control group(35.64±5.86 vs 29.45±7.04, P<0.05). On day 3, the two groups was no significant difference (P>0.05). On day 1,2 after PH, the mRNA and protein expression of Cyclin Dl in UDCA group was significantly higher than the control group(P<0.05), and no significant difference between the two groups on day 3.Conclusion:Ursodeoxycholic acid can promote liver regeneration, and increase the expression of cyclin D1.
