Objective:Comparative analysis of experimental rats miniature planting of anchorage microimplant (served) bone healing process and normal rats bone apoptotic gene B c1-2 and B ax protein expression of significance and difference. A preliminary discussion the role of apotosis in healing of implant.Methods:40 SD rats were randomly devided into three groups:groupA(contral group),groupB (implant group),groupC(model group). Model group (SD) rats were injecteed SIZ to establish diabetes model.Glucose values≥16.7mmol/L are successful. Then randomly divided into the diabtes group (C, n= 10) and the diabetes with implant groups (D, n=10).Then 40 SD rats were feeded after two weeks in room-temperature. Estabish implant geoup and diabetes with implant group.Then randomly extracting the side of shin implant microimpla anchorage. After eight weeks killed all rats. Take the bone slices which were including Microimplant anchorage.The bone slices were analyzed by immunohistochemistry fou bcl-2 and bax,TUNEL methods fou apoptosis,HE fo Morphological organizationResult:(1)General observation:ALL experimental animals were no postoperative inflammation and no sudden death happended. Check up the stability of implant regulary,the implant stabilization of each stage were very well. The organization around the implant was no flare, and the color were very well also.(2) The results observed by HE shouwed as follows:At 1 weeks,there wsa inflamation at the interfere,appear some osteoclast,nucleus are increase obviously a few of collagenous fiber tissues at the interfere. At eight weeks,bone collagenous fiber tiss,bone fossa,bone trabecula and some osteoblast around the bone trabecula were seen at implant interfere.(3)The TUNEL check:The apoptosis tates of planting in group C (26.38±2.67%)and D (17.37±2.15%) were incressed compared with that of A(5.5±1.65%) group and B (5.2±1.37%)group. Significant different of apoptosis rate were found betwen contral group and Microimplant anchorage group.(p<0.05). Significant different of apoptosis rate were found betwen C group and D group (p<0.05). No significant different of apoptosis rate were found between A group and B group(p>0.05). (4) the expression of Bcl-2 protein in bone cell:The expression of bcl-2 in bone cell both in group A(37.86±2.56%) and group B(37.23±3.01T%). No significant different of apoptosis rate were found between A group and B group(p>0.05). The expression of bcl-2 in bone cell in group C (50.13±3.01%)and D (59.87±3.47%) were incressed compared with that of A and B group. Significant different of apoptosis rate were found betwen contral group and Microimplant anchorage group.(p<0.05). Significant different of apoptosis rate were found betwen C group and D group (p<0.05).(5) he expression of bax protein in bone cell:The expression of bax in bone cell both in group A(37.80±1.75) and group B(38.00±1.23). No significant different of apoptosis rate were found between A group and B group(p>0.05). The expression of bcl-2 in bone cell in group C (55.78±4.89)and D (48.98±4.10) were incressed compared with that of A and B group. Significant different of apoptosis rate were found betwen contral group and Microimplant anchorage group.(p<0.05). Significant different of apoptosis rate were found betwen C group and D group (p<0.05).(6)The ratio of the positive rate of bcl-2 expression to that of bax:No Significant different of apoptosis rate were found betwen C group and D group (p>0.05).(7) Spearman correlation analysis shouwed that bone cell apoptosis rate was negatively correlated with bcl-2/bax ratio (p<0.001) and the correlation coefficient was-0.523.Conclusion:1.The apoptosis rate are increase in diabetes rats with implant, show the apoptosis are relevant with osseointegration reduce which under the condition of diabetes.2. Bcl-2/bax reduce, shuw bcl-2和bax are participate the apoptosis which diabetes rats wthu implant.
