Objective:To study the preparation and properties of Transforming growth factorβ1(TGF-β1) gelatin microspheres.Evaluate the biological effects of TGF-β1 gelatin microspheres on the proliferation of human periodontal ligament cells and confirm whether controlled releasing system is appropriate for treatment of periodontal disease.Method:TGF-β1 gelatin microspheres were prepared by improved emulsified cold-condensation method.The morphology of microspheres were observed with SEM and tested the encapsulation rate,drug content and the properties of drug release;Colorimetric MTT method were used to observe the proliferation of HPDLCs after different doses (lmg/ml,5mg/ml,10mg/ml)of TGF-β1 gelatin microspheres were added into the cultrue medium.Result:The diameter of gelatin microspheres were 10.28±2.24μm in average.The encapsulation rate was 78.5%, and drug containing was 785ng/g,94%of the drug was sustained-releasing within 7 days. TGF-β1 promoted the proliferation of HPDLCs obviously.Conclusion:The preparation of TGF-β1 gelatin microspheres is easy and it can promote the proliferation of HPDLCs through a long period of controlled release of TGF-β1. TGF-β1 gelatin microspheres may be an promising durg dose be used to treat periodontal disease.
