Metal-ceramic alloys are widely used in clinic nowadays, but affected by the saliva the metal ceramic alloys in the patient's mouth will release some metal ions which may cause the gingivitis or even discoloration of the gingival tissue. It's important to investigate the biocompatient of the metal-ceramic dental alloys.This research were divided into two parts:firstly MTT assay was used to measure the cytotoxicity of three Nobel-ceramic alloys which were usually used in the department of prosthetics in Stomatology hospital of Shanxi medical university to compare the activity and proliferation on mouse fibroblast L-929.Then ELISA was used to detect the secret of IL-6 on L-929 to study the influence about immunity function of L-929.Objective:To evaluate the cytotoxicity of three kinds of noble ceramic alloy (89%Au-alloy/ 74%Au-alloy/Ag-Pd-alloy) in vitro and to investigate the assays about how to test the bio-safety of oral biomaterials. Methods:Mouse fibroblast L-929 were cultured with extracts of these three kinds of noble ceramic alloys. The cyto-activity in cultured L-929 were assessed by means of Tetrazolium-based colorimetric assay on 24h/48h/72h respectively and ELISA was used to detect IL-6 secretion of the supernatant of L-929 on 24h and 48h. Results:(1) The cyto-activity of L-929 which were cultured in these three noble ceramic alloys were no significant differences with that of negative group, and they all showed their better bio-safety compared with the positive control group. But multiple comparisons indicated that the cyto-activity which was cultured in 89%Au-alloy was least among these three kinds of materials in 72h. (2)The IL-6 level of the supernatant in the Ag-Pd alloy and the positive control group were statistically lower than that in negative group on 24h, and which were lower than that of 48h means that Ag-Pd alloy was similar with the positive control group in this point. Conclusion:The results of the present study suggested that different Nobel-ceramic alloys be different to cells. The cytotoxicity of materials be not ratio to the golden contents, maybe relate to all ingredients or different technology of factory.74%Au-alloy was the best material among these three kinds of noble ceramic alloy.
