Purpose1. Research the activity of pancreas nuclear factor-κB (NF-Kb) on normal rats and the quantity of serum tumor necrosis factor (TNF-a).2. Research the change of the pancreatic tissue NF-κB activity and serum TNF-a amount in acute pancreatitis (AP) rat, explore the function of NF-κB and TNF-a in rat pancreatic injury, and clearly explicit the part of the mechanism of pancreatic injury when AP.3. Research the change of NF-κB activity and serum TNF-a amount after application of arginine (L-Arg) in AP rats, clear therapeutic effect of arginine on the AP to explore the mechanism of arginine on the treatment of AP.4. Research the change of NF-κB activity and serum TNF-a amount after application of leptin (LEP) in AP rats,clear therapeutic effect of leptin on the AP to explore the mechanism of leptin treatment on the AP.5. Research the change of NF-κB activity and serum TNF-a amount after application of arginine combined leptin in rats, clear the therapeutic effect of arginine combined leptin on AP whether superior to single drug treatment, explore the mechanism of arginine combined leptin to provide a theoretical basis for treatment of AP with arginine combined leptin.Materials and methods1. Wister rats 60, were randomly divided into sham operation group (A group), acute pancreatitis group (B group), L-arginine treatment group (C group), leptin treatment group (D group), mixed-treatment group (E),12 rats in each.2. acute pancreatitis model was made by retrograde injection of 5%sodium taurocholate into cholangiopancreatography. At 6 hour of the model, anesthesia, Each group of rats were did intraperitoneal injection drug before closed abdominal, then get Abdominal aortic blood and pancreatic specimens; detect tumor necrosis factor-a(TNF-a) levels, serum amylase, nuclear factor-KB (NF-κB) activity of pancreatic tissue, and evaluate pancreatic tissue Pathological damage score in each group. Results1. Sham operation group, no pathological damage of the pancreas, organizations NF-κB, serum TNF-a and amylase were only a weak expression2.The pancreatic damage of AP group was clear and NF-κB, serum TNF-a, amylase were significantly higher than sham operation group (P<0.05).3. The pancreatic damage of arginine treatment group was reduced, NF-κB, serum TNF-a and amylase were lower than AP group (P<0.05).4. The pancreatic damage of leptin treatment group was reduced, NF-κB, serum TNF-a and amylase were lower than AP group (P<0.05).5. The pancreatic damage of Mixed treatment group was reduced, NF-κB, serum TNF-a and amylase were lower than arginine group and leptin group (P<0.05). In each group, the NF-κB and serum TNF-a were positively correlated (P<0.01).Conclusion1. NF-κB Exists in normal rat pancreatic tissue and the activity is not high, serum TNF-a is in low level.2. pancreas was obviously damaged and pancreatic tissue NF-κB, serum TNF-a were significantly increased in AP, and both were positively correlated, indicating the occurrence of AP associated with the increas of NF-κB activity and over-expression of TNF-a, NF-κB promoted TNF-a production.3. After application of L-arginine or leptin, pancreatic damage was reduced, NF-κB and serum TNF-a were reduced, indicating both can cut down the activity of NF-κB so that the production of TNF-a was reduced, It has obvious therapeutic effect on the AP.4. After application of L-arginine combined leptin, pancreatic damage was significantly reduced, NF-κB and serum TNF-a were more reduced, indicating the two joint can enormously cut down the activity of NF-κB, the two-drug combination treatment was superior for single-drug therapy.
