Objective: To study the effects and mechanisms of ursolic acid on human pancr- eastic carcinoma cell line PANC-1 and to investigate the expression of CD-105 and D2-40 in the pancreatic ductal adenocacinoma(PDAC).Metheods: After UA treatment, the growth inhibition of PANC-1 cells were assessed by MTT assay. Cells were evaluated with flow cytometric analysis, Hoechst 33258 staining and transmission electron microscope after it was induced by UA;It's mechanisms was determined by RT-PCR analysis. Using immunohistochemical staining and tissue microarrays, expressions of p53,EphA2,VEGF,CD105 and D2-40 were examined.Results: The proliferation of PANC-1 cells were significantly inhibited in a dose-and time-dependent manner after UA treatment. Cell apoptosis ratio and cell S phase arrest was examined by FCM; After cells were treated by UA(40μmol/L), the expressions of p53 were increased whereas VEGF and EphA2 decreased. The immunohistochemical staining show that expressions of p53,EphA2 and VEGF in tumor tissue were significantly higher than in the non-tumorous pancreatic tissue. Possitive relevances were found between CD105-MVD and p53,EphA2 and VEGF. Possitive relevances were also found between D2-40-LVD and p53 or EphA2.Conclusions: UA can induce cell apoptosis. These effects might be related with the expressions of p53, the expression of p53 related to the expressions.UA can decrease the expression of EphA2 and VEGF, then UA suppress growing of blood vessels and lymthatic vessels of tumors.
