Background:Oral candidiasis is caused by candida species infections.In recent years,with the wideness use of broad-spectrum antibiotics,cytotoxic drug and immunosuppressive drugs,the increase of AIDS,diabetes,cancer patients, the evolve of many therapeutic tools such as organ transplantation and catheter intervention, the host immune function is low or dysbacteriosis,so that the incidence of oral candidiasis is rising rapidly. Although candida albicans are the most important pathogenic fungus when the conditions are siitable,the strain of weaker virulence infections as candida krusei is repeatedly happen in immunocompromised patients.The influence of oral candidiasis pathogenesy not only related with the morphous of thalline itself,surface structure and bioadhesion,but also concerned with the host's immune defenses. IL-8 is neutrophils powerful chemotactic factors, in the process of immune regulation and inflammation plays dual role. Plenty of domestic and foreign research shows that in the oral candidiasis, candida albicans can stimulate oral mucosal epithelium cells to produce cytokines such as IL-8, participate in the mucous local defence reaction,however the ability of candida krusei adherence to oral epithelium cells and induce IL-8 responses as single infecting organism and in combination with Candida albicans was seldom reported, the exact mechan(?)relevant IL-8 with oral candida infection is not very clear.Objective:The aim of the study was to discuss the adhearance and differences between Candida albicans and Candida Krusei,compare the expression of interleukin-8 of oral epithelial cells under the stimulate of Candida Krisei single and mixed infection,to investigate the possible effection of oral epithelial cells defense candida infection. Methods:1.Put slides into cell culture capsule,and then vaccinated KBcell to the culture capsule to cultivante 48 hours,after contact with the candida suspension of candida albicans and candida krusei for 1.5,3,4.5,6,24 hours,took out the slides and fixed with absolute alcohol,microscopically counting the adhesion of candida on epithelial cells after gram stain; After collecting trypsin digestion and monilia, the rejection experiment to detection candida vitality and conversion hyphae 2.Coculture KB cell with phase form and hypha form candida suspension of candida krusei and mixed with candida albicans and for 6,12,24,48 hours,unstimulate cells as positive control. At the end of each contact period,the culture supernatants were harvested,IL-8 protein levels were analyzed by enzyme-linked immunosorbent assays (ELISA).3.Tolal cellular RNA was abstraction by Trizol when two species candida cocultured with KBcells for 24 hours,IL-8mRNA expression was determined by reverse transcription polymerase chain reaction (RT-PCR).The t test was used to analyse the experimental data to judgement the statistical significance.Results:1.The adherence of candida to KB cell have tine dependence, along with the time extend,the adherence was gradually increased,the adherence of candida krusei was lower than candida albcans.;The transformation of candida from blastospores to hyphae was time dependent.the percentage of the hyphal form rose as time extend,but the sum of candida were reduced.2. The level of IL-8 of experiment group was higher than control group The expression of IL-8 was also time dependence, achieve a peak at 24 hours and then appeared downturn.,and the expression of candida krusei was higher than mixed infection.3.KB cell was coculture with two species candida for 24 hours,the expression of IL-8mRNA was higher than control group,and the candida krusei was higher than mixed infection.Conclusion:1.The adherence of Candidiasis to oral epithelial cells was a kind of specific adhesion,the ability of Candida krusei adherence was lower than Candida albicans.2.The form of blastopores transform to hyphae can help Candidisis escape epithelial cell inhibitory effect,which have a relationship with the pathogenicity. 3.Candida krusei is responsible for the production of IL-8 secreted by oral epithelial cells which can be expected to play an important role in the infection and defection with the local oral mucosa.4.The low expression of Candida Krusei mixed with Candida albicans indicates that antagonism between Candida albicans and Candida Krusei.
