IntroductionLeptin derived from the Greek, also has been known as slim factor, discovered by Zhang first in 1994. Leptin is a kind of obese gene (OB) encoding peptide hormone that contains 146 amino acids, molecular mass of 16 kilodalalton(KDa), produced mainly by adipose tissue, is multi-target organs, a wide range of features protein hormones.1998 Potter, etc. in the activation of hepatic stellate cell(HSC) found the leptin mRNA and protein, people started to look at the relationship between leptin and liver disease. In recent years, many research found that leptin and the occurrence development,clinical presentation and prognosis, etc. of chronic liver diseas has more closely related. Domestic and international study found that the performance of leptin in patients with liver cirrhosis abnormalities, but the result reported different.Liver cirrhosis is a kind of characterized by the formation that diffuse hepatic fibrosis and regenerative nodules pseudolobule chronic liver disease, common disease, common disease in our country, and one of the causes of death, our patient with liver cirrhosis accounted for the total number of internal medicine 4.3%~14.2%,so in-depth study of serum leptin and the intrinsic link between liver cirrhosis with the clinical significance can not be ignored.The topic observed and compared serum leptin levels on the liver cirrhosis patient and healthy controls, and analyzed the affecting factor of leptin in patients with cirrhosis and investigated the mechanism of occurrence and development that serum leptin on liver cirrhosis.Materials and methodsI. Sample Collection1. Liver cirrohosis:Selecta a total of 120 case of the cirrhosis of the liver cirrhosis that in 2008-2009 hospitalized in our hospital gastroenterology. male 90 case, female 30 case, according to child-pugh grade:A grade-36 case, B grade-42 case, C grade-42 case.2. Normal group:Select 20 case the health of those who were proven by medically in 2009, and control their sex, age, BMI approximately match with the liver cirrhosis. 13 patients were men and women in 7 cases.II. Test Method1. Blood samples obtained:subjects caught more than 8 hours fasting, early morning bleed from venous and collected 3ml, in 2 hours centrifuged the serum, preserved them in-70℃refrigerator to wait for detecting leptin.2. Human body paramenters measured:recorded the age, sex, height, weight of subjects, and calculated body mass index(BMI), BMI-weight/height2 (kg/m2).3. Detection of serum leptin:serun leptin was determined by Wuhan Boster Engineering Co., Ltd. provided leptin kits, using ELISA, in strict accordance with the instructions to operate.4. Statistical methods:data was statistically analyzed by sigle-factor analysis of variance and Q test (SNK) with spss 11.5 software. Date was expressed by mean±standard deviation.Results1. Subjects age and BMIIn liver cirrhosis group and control group, the groups of liver cirrhosis with different liver function, the subjects'age and BMI compared with no statistical difference.2. Different liver function of liver cirrhosis serum leptin levels①Serum leptin levels in patients with liver cirrhosis were significantly higher than control group (p<0.01).②In the different group of liver cirrhosis liver function of the child-pughA, B, C, the serum leptin levels of any one group was significantly higher than control group (p<0.01,p<0.01,p<0.01).③With the gradual deterioration of liver function, serum leptin levels showing progressive increase trend, at the same time, serum leptin levels of child-pugh C-level was significantly higher than child-pughA-level (p<0.01). 3. Different genders patients of liver cirrhosis serum leptin levels①Both liver cirrhosis group and control group, women leptihn were significantly higher than men's (p<0.01,p<0.01).②In the same group of liver cirrhosis liver function child-pugh gradeA, B, C, women leptin were always significantly higher than men's(p<0.05,p<0.05,p<0.01).Conclusion1. Serum levels in patients with liver cirrhosis compared with healthy control group significantly increased.2. Serun leptin levels in patients with liver cirrhosis is closely related to child-pugh grade.3. Serum leptin levels is exist of gender differences.
