ObjectiveWe successfully product the rats model of chronic interstitial nephritis (CIN) induced by adenine and then injected the mesenchymal stem cells(MSCs) into the rats by tail vein in vivo in order to further evaluate mesenchymal stem cells in the treatment of animal models of chronic interstitial nephritis.MethodsMethods:We fed the rats with adenine (100mg/kg/d) everyday for about 4 weeks and stopped feeding for 4 weeks.After that, we dected the clinical manifestation,biochemistry in order to judge the animal model.During that period, we successfully stripping the femur, removed the bone marrow to the cell suspension, isolated and cultured in a Petri dish. When the cell were adherent, confluent, just like the spindle, we passaged the cells,3 generationgs back-pass, then used immunohistochemistry to identify cells CD90,CD34 expression.When we were sure the animal model and cells culture successful, we divided the remaining rats into three groups(A,B and C).A group (transplanted group) were injected with MSCs, B group (non-transplanted group) with normal saline, C group (control group) were not interfered with anything. We observed the clinical manifestations,weight changes of rats every day and till the 4 weeks we killed the rats and examined the clinical scores,renal function as well as the pathological changes of kidneysResultsThe animal model:The adenine fed rats showed significant weight loss, polydipsia and polyuria, hair loss, low spirit,convulsions and even death. We made a clinical score with clinical manifestation and compared with creatinine in order to judge the clinical score.We found that there was a significant correlation between creatinine and clinical score and deduced a success about the clinical score table.Compared with the control group, there was a significant difference (P<0.05) about clinical score, biochemical and pathological score and the pathological manifestations of kidney were mainly on renal tubules, interstitial damage,the glomerulus were changed very little. So we got the conclusion that the animal model were successful. Mesenchymal stem cells:The cells can be seen to plastered bottom, changed the formation of spindle cells, some cells were confluenced and express CA90+, CD34- determine mesenchymal stem cells successfully.After injected with stem cells,we compared the three goups with each other. There were different degrees of deterioration on the transplanted group and non-transplanted group about the clinical manifestations, biochemical and pathological findings.There was significant statistical difference among the three groups (P< 0.05).Howerer when we compared with each group,we found that there was no significant difference (P> 0.05) between the transplanted and the non-transplanted group.ConclusionThe rat model induced by adenine was successful. However we failed to achieve the expected results,and Mesenchymal stem cells can not reverse the chronic interstitial nephritis and its pathological changes, and played a limited role in the treatment of chronic interstitial nephritis.
