ObjectiveTo study the effect of local injection of different concentrations Pingyangmycin in rabit lip muscle tissue and detect the expression of bFGF and VEGF in muscle, lip hemangioma treatment for clinical application of PYM provide experimental and theoretical basis.MethodsUse 6 to 8 months,28 healthy rabbits, weighing 2.0-2.5kg, no restrictions on gender. In which 24 rabbits as experimental group,and were randomly divided into A, B, C three group, n=8, A group of high concentration group (2.7mg/ml), B group for the middle concentration group (1.6mg/ml), C group with low concentrations (1mg/ml), the other 4 rabbits as the saline control group (D group). By design, the concentration of each group, respectively 1 day and the first eight days on the bilateral upper lip intramuscular injection of 0.5ml of PYM, at a distance for the first time 28 days later injected area injected muscle tissue biopsies and pathological examination, observe the morphological changes, and used immunohistochemistry to detect the expression of bFGF and VEGF in muscle tissue.Results1. Histological observation:Experimental group, pathological changes in lip muscles can be seen in the A Group shows muscle fiber bundles were irregularly arranged, showing ripple-like, stripes and a little fuzzy, a small amount of condensation nucleusi, eosinophilic change, swelling, fusion, B group shows muscle fiber bundles eosinophilic change interval, muscle fiber bundles is loose, eosinophilic change, fusion, C group muscle fibers in Closely arranged, sarcolemma integrity, muscle fiber bundles showed eosinophilic change interval, degeneration of muscle fiber bundles with the trend of convergence. D group showed a normal lip muscle histological structure, tightly packed muscle fibers, size, stained uniform, Small nucleus, located at the cell edge, rules of light and dark stripes on the muscle fibers and tidy, sarcolemma integrity, and clarity.2. the expression of BFGF:D group of bFGF expression in lip muscle can be seen, A,B group the expression of bFGF group was significantly higher than D, the difference was statistically significant (P<0.01), the expression of bFGF in C group, although there is an upward trend, But the Cgroup and D group difference was not statistically significant (P> 0.05), With the increase of drug concentration in the experimental group gradually increased the expression of bFGF, A, B, C groups compared with the three groups was statistically significant (P<0.05).3. the expression of VEGF:Group D was almost no VEGF expression in lip muscle, A group, B group and C group of VEGF expression in lip muscle can be seen, the above three groups as compared with the D group was statistically significant (P <0.01), A, B, C among the three groups in each group compared with no statistical significance (P> 0.05).Conclusions1. Injection of PYM can lead to irreversible lip muscle damage, the higher the concentration of PYM injections, leading to more severe damage.2. Caused by injection of different concentrations of PYM lip muscle changes in the expression of VEGF and bFGF is one of the important reasons for lip muscle atrophy, degeneration.
