Relationship Between TP,VEGF Expression And Hepatocellular Carcinoma Angiogenesis

ObjectiveBy detecting the different expression levels of TP,VEGF,MVD in paraffin-embedded clips of 72 cases of hepatocellular carcinoma(HCC) and 19 cases of normal liver tissue,analysis the different expression levels of them in HCC tissue and normal liver tissue;analysis the relationship between them and Edmondson class,tumor size,integrity of tumor envelope and TTPV;investigate whether TP and VEGF have synergism in HCC angiogenesis,and whether they can provide a new way to the clinical treatment.Method1.Samples(1)HCC group:collect 72 paraffin-embedded clips of HCC(without radiation or chemotherapy before operations),which were obtained by operation in Shengjing Hospital of China Medical University from January 2008 to March 2009.Among the HCC group,included 50 cases of male,22 cases of female;average age is 55.6±7.2 years old.(2)Control group:collect 19 paraffin-embedded clips of normal liver tissue in hepatic benign tumor,included 12 cases of male,7 cases of female,average age is 42.9±5.4 years old.2.MethedApplying SP immunohistochemical stain technique,detect the different expression levels of TP,VEGF,MVD all of the samples.To determine the result according to positive contract clips. ResultTP,VEGF are mainly expressed in cytoplasm of the HCC cells;CD34, which marked in vessel,is mainly expressed in stroma.TP expression in HCC group,the rates of strong positive,weak positive and negative are 28 cases(38.9%),28 cases(38.9%) and 16 cases(22.2%); in benign group, rates are 3 cases(15.8%),6 cases(31.6%) and 10 cases(52.6%).What's more,VEGF expression in HCC group,the rates of strong positive,weak positive and negative are 33 cases(45.8%),23 cases(31.9%) and 16 cases(22.2%);in benign group,rates are 3 cases (15.8%),5 cases(26.3%) and 11 cases(57.9%).MVD mean is 19.34±1.21 pieces/HP,4.23±1.66 pieces/HP separately in HCC group and benign group.There are apparent variabilities of TP,VEGF,MVD expressions between HCC group and benign group(P<0.05).In HCC group,cases of both TP and VEGF expression positive,MVD mean is 24.75±3.22;cases of whether TP or VEGF expression positive,MVD means are 14.35±4.00,11.65±3.14;cases of both TP and VEGF expression negative,MVD mean is 4.47±2.60.There are apparent variabilities between both positive group and simple positive group,as same as between both positive group and negative group(P<0.05);but there is no apparent variability between simple positive group and negative group(P>0.05).In this research,expressions of TP,VEGF have apparent relationships with Edmondson class,tumor size,integrity of tumor envelope and TTPV(P<0.05).ConclusionThere are apparent variabilities between both positive group and simple positive group,as same as between both positive group and negative group;but there is no apparent variability between simple positive group and negative group.TP and VEGF have synergism in HCC angiogenesis.
