Low molecular weight heparin is a mixture extracted from unfractionated heparin by the methods of Hydrolysis or chemical degradation.The relative molecular mass of it is 3-8 KDa, and the average is 5 KDa.Compared with unfractionated heparin, low molecular weight heparin has the following advantages,such as well subcutaneous absorption, longer half-life, bioavailability, good homogeneity, strong anticoagulant effect,obvious effect of hypolipidemic, less side effects, without laboratory monitoring and so on.Low molecular weight heparin modified by chlorosulfonic acid– pyridine is called polysulafted heparin because of the introduction of sufate in it.Its relative molecular weight is 6-13 KDa.This paper choosed low molecular weight heparin as experimental material to try to have an analysis of its antibacterial, tumor suppressor and lipid-lowering activity.⑴in vitro antibacterial effects of LMWH This paper used agar diffusion and 2-fold dilution methods to test the in vitro antibacterial effects of low molecular weight heparin and polysulafted heparin on Staphylococcus aureus and Escherichia coli.The results showed that: Both heparin polysaccharides had inhaibitory effects on the common Gram-positive and Gram-negative bacteria;The differences between the inhaibitory effects of low molecular weight heparin and heparin on Staphylococcus aureus were significant,but there was no significant difference between the inhibitory effect on E.coli;The results of the minimum inhibitory concentration experiment showed that the inhibitory degree of low molecular weight heparin and polysulafted heparin on Escherichia coli was stronger than that on Staphylococcus aureus.The tests of detemining drug sensitivity showed that low molecular weight heparin and polysulafted heparin ware moderately sensitive to drugs and had better inhibitory effects on Gram-negative bacteria.⑵The inhibitory effects of proliferation in vitro of low molecular weight heparin on four human cancer cell.This paper uesd cell passaged culture to culture cancer cells and MTT method to measure the absorbance value and growth inhibition rate of cells.It inverted microscope to observe changes of cell morphology.And It applied flow cytometry to test cell cycle and cell apoptosis. And a dose-dependent and time-dependent manner, G0/G1 phase cells increased, S phase cells decreased,The results showed that the inhibitory effects of proliferation of low molecular weight heparin on gastric cancer cells (SGC-7901) are significant. G0/G1 phase cells increased, G2/M phase and S phase cells decreased,but it could not induce apoptosis in gastric cancer cells.Low molecular weight heparin can reduce the lung cancer cells (A549) activity. In the G1 phase ratio increased significantly,but a significant decrease in S phase, But it does not increase the percentage of dead cells. Low molecular weight heparin could inhibit the proliferation of human breast cancer cells (MCF-7).Use MTT method to test the growth inhibition of low molecular weight heparin on hepatoma cells (HCCLM3) cultured in vitro.The results show that the inhibition was great and it increased in accountance with time and concentrations.Cell apoptosis rate was 48.43 percent.⑶Heparin blood lipid test compounds.This experiment used the high fat diet formula fed inducing hyperlipidemia in mice model and measured its blood parameters.The results showed that compared with hyperlipidemia mouse model group, different doses of heparin group had cholesterol levels decreased in varying degrees,and also showed that the middle-dose low molecular weight heparin group and the high-dose group could significantly reduce blood cholesterol in hyperlipidemia small mouse.The results also showed that high-dose low molecular weight heparin could reduce serum cholesterol hyperlipidemic mice up to 42.72%;and the MDA levels in middle-dose and high-dose low molecular weight heparin groups ware lower than that in the model group.It explained that low molecular weight heparin could enhance antioxidant capacity.Conclusion: Both low molecular weight heparin and heparin can inhibit the growth of Gram-positive and Gram-negative bacteria and both of them are medium-sensitive drugs.Low molecular weight heparin can inhibit the proliferation in vitro of four types of human cancer cell,can arrest cell cycle in G0/G1 phase,and can induce apoptosis,but it can not induce apoptosis in human gastric cancer cells;The middle-dose low molecular weight heparin group and the high-dose group can significantly reduce Serum cholesterol and MDA content in hyperlipidemia mice.
