Effect Of Compound "Tong Gu Luo" On Proliferation Of Human Osteosarcoma Cell Line MG-63

Osteoporosis is a systemic skeletal disease characterized by low bone mass and microarchitectural deterioration of bone tissue,leading to increase on bone fragility and a risk of fracture. It is a commom and frequently occuring disease among the middle-aged and elderly people.Osteoblasts are the main functional cells in bone formation and, therefore, agents stimulating osteoblastic bone formation would have potential activity of anti-osteoporosis. In order to exploit novel and effective medicines to prevent and cure osteoporosis,osteoblast-like MG- 63 cells were employed as a modle of drug efficacy appraisal. The cell proliferation was measured as index of stimulating osteoblastic bone formation and assayed by MTT method.Compound "Tong Gu Luo" is used to treat osteoporosis as a prescription of traditional Chinese medicine, which is composed of four herbs such as Polygonum multiflorum , Cuscuta chinensis Lam and other two Chinese medicines. In order to determine the active components of each herb in Compound"Tong Gu Luo", the crude extracts of every herb were extracted by ultrasonic extraction method. Ethyl acetate, 95% and a certain concentration of ethanol solution were used as extract solvent. Three parts were obtained from each crude extract. At last, twelve parts (H₁, H₂, H₃, T₁, T₂ ,T₃,A₁, A₂, A₃, B₁, B₂ and B₃) were obtained.In order to determine the superiority concentration of every herb in compound"Tong Gu Luo", according to orthogonal test design, the influences of combination extracts in the cell proliferation rate of MG-63 were observed and studied by MTT assay. The best combination is: 0.2mg/mL of Polygonum multiflorum,0.4 mg/mL of Cuscuta chinensis Lam,0.4 mg/mL of A and 0.2mg/mL of B.In addition, the effect of twelve components in their respective superiority concentration on the proliferation rate of osteoblast-like MG-63 cells was also studied. The experimental results show that the function of which promote cell proliferation is from strong to weak respectively is: A₂→B₂→H₂→A₁→T₁→A₃→B-3→B₁→T₂→T₃→H₃→H₁This thesis, the effect of Compound"Tong Gu Luo"on proliferation of human osteosarcoma cell line MG-63 was observed and studied. The efficacy of this prescription of traditional Chinese medicine on promote cell proliferation was evaluated. The research had laid the foundation for identifying the effective constituents of Compound"Tong Gu Luo"to cure osteoporosis and establishing quality standard of Compound"Tong Gu Luo".
