Objective:To study the anticancer metastasis compounds's mechanisms of anticancer, we took transplanted S180 cancer of mice as target to study the mechanisms of Anticancer Metastasis Compound (JB and SDG).Methods:Estabish the S180 tumor-bearing mice model, we observed inhibition ratio of three doses of JB and SDG. To analyze effect of JB and SDG on prolonging tumor-bearing mice's life span. We studied the effect of JB and SDG on immune function of tumor-bearing mice by detecting splenic weight, splenic index, thymus weight, thymus index and T cell subset. We studied the side effect of JB and SDG on tumor-bearing mice by detecting liver function .We also studied the expression of TNF-αand CD34 in each group to analyze the effect of JB and SDG on tumor angiogenesis.Resluts:1. The inhibition ratio of middle dose of JB and SDG in SDG group is 62.93%, in JB group is 67.28%, close to 5-FU group 26.73%. The inhibition ratio of high dose of JB and SDG in JB group is 26.73%.2. The splenic weight and splenic index of the low and high dose of SDG and JB groups were higher than that of 5-FU groups correspondingly. Although the difference did not reach statistically significant level, but the increasing trend is obvious.3. JB and SDG administration for 18 days, compared with model group, albumin levels of SDG groups were near the control group, and significantly higher than the 5-FU groups (P <0.01). Albumin levels of 5-FU, low and middle doses of JB groups decreased significantly. ALT levels of treated groups all decreased and AST levels had no significant changes. Administration for 25 days, compared with model group, Albumin levels of 5-FU groups and SDG groups of treated groups no significant difference; ALT and AST levels of treated groups had no significant change. Administration for 46 days, liver function of 5-FU groups and SDG groups had no significant change.4. JB and SDG has the effect on increasing the expression of CD34, but 5-FU group increased obviously (P<0.05).5. Contrast with model control group, treated groups significantly prolonged tumor-bearing mice's life span except low doses of JB groups. SDG groups prolonged obviously (P<0.01)and the rest of groups prolonged life span too(P<0.05). The ratio of SDG groups'prolonging life span is 108.04%, higher than 5-FU groups.Conclusions:1. All three doses of SDG and JB groups have inhibitory action on S180-liver tumor-bearing mice. It has obviously time-effect relationship.2. SDG groups and JB groups have the effect of enhancing immunological function of tumor-bearing mice. This effect of low doses of JB groups is obviously better than 5-FU groups.3. SDG groups and JB groups have no significantly damage to liver function.4. SDG groups and JB groups have the effect of enhancing the expression of TNFα, and it may be related to suppress amplification and metastasis of tumor.5. SDG groups and JB groups have the effect of prolonging life span of S180-liver tumor-bearing mice and the ratio of SDG groups'prolonging life span is 108.04%, higher than 5-FU groups.
