Objective: By studying the expression level of GRP78 protein in primary multifocal hepatocellular carcinoma tissue sources of different clones, intrahepatic metastasis (IM) group and multicentric occurrence (MO) group and a single nodule (S) group and the normal liver (N) group,To explore the invasion and metastasis correlation in different clones of primary multifocal hepatocellular carcinoma.Methods: Immunohistochemistry, RT-PCR, Western blot was used to detected 17 cases of IM group,21cases of MO group,8 single-nodular HCC tissues and 14 normal liver tissues in the relative expression of GRP78.Results:According to the results of western blot GRP78 expression level in IM,MO,S,N tissue are reduced(p <0.05); According to the results of RT-PCR GRP78 expression level in IM,MO,S,N tissue are reduced(p<0.05); According to the results of Immunohistochemistry The positive rate of GRP78 in IM group is 60.0%,47.7% in MO group,62.5% in single nodule group,The intensity of positive signals represented by OD value,more than 30% considered as positive,less than 30% considered as negative.There are a significant difference shows in the positive rate of IM group compare with that of the MO group (p=0.002).Conclusions:There are differences of GRP78 in multifocal hepatocellular carcinoma tissue sources of different clones,as well as normal liver tissues.Over-expression of GRP78 protein may be related to multifocal hepatocellular carcinoma occurrence,development,biological behavior and prognosis.it may serve as a prognostic biomarker to distinguish IM and MO.
