Background Head and neck cancer remains one of the most devastating cancers, and nearly 90%of these cancers are squamous-cell carcinomas. Head and neck squamous cell carcinoma (HNSCC) is the sixth most common cancer with an annual incidence of approximately 400,000 worldwide. Genomic DNA of oncogenic Human papillomaviruses (HPV) is detected in approximately 26% of HNSCC worldwide. Recent Studies have shown that HPV infection associated with relatively subtle genetic alterations in HNSCCs. studies show that infections with high-risk (oncogenic) types of HPV is play an important role for in the development of HNSCC.Purpose To study the interrelationship between the HNSCC and HPV in Sichuan province of China, and describe the distribution, expression of genotypes of HPV in the HNSCC. Study its the role in the development of HNSCC. Method The HPV genotypes in 45 HNSCC specimens were detected by a combination of MY09/11 consensus primers PCR (MY09/11 PCR), type-specific primers one-step PCR (One-step TS PCR) and E6/E7 gene type-specific primers nested PCR (Nested TS PCR). The prevalence and distribution of HPV in HNSCC patients with especially for HPV types 16, 18,33,45,52, 58 and 59, suspected to be most common in all world, was investigated.The majority of tumors were located in the oral cavity(18 cases), oropharynx(6 cases), larynx(19 cases), nasopharynx(2 cases).Results Of the 45 HNSCC patients, 7 (15.6%) had detectable HPV DNA in their tumor samples. HPV DNA was detected 2 of 19 larynx tumors, 1 of 6 oropharyngeal tumors, 3 of 18oral cavity tumors and 1of 2 nasopharynx tumors. And all of them cases are infected with HPV16. One of them infected with both HPV16 and HPV52 viruses. Sequencing analysis revealed that the mutation of the case of HPV16 type L1 genes 6224 c→g, 6930 a→c had not been reported.Conclusion Preliminary investigations reveal that the HPV infection rate of HNSCC in the Sichuan province low than othei areas. HPV16 is majority in HPV-type .6 cases of integrarion were detected in 7 cases of viral oncogenes show the high rate of integrarion in HNSCC. One case of HPV L1 mutant strain belong to Europe strain which was found through the evolutionary tree. Our study indicated that the rate of HPV infection are varying in different sexual behavior ,and are varying in different countries and different areas of the same country .
