Background and object:Hepatocellular carcinoma (HCC) is one of the highest incident malignant tumors in China. A lot of factors were induced the HCC. Autophagy is an evolutionary conserved pathway that involves the sequestration and delivery of cytoplasmic material to the lysosome, where it is degraded and re-cycled. Some studies have indicatedthat the down regulation of autophagy-related genes might result in tumorigenesis. This study was to investigate the expression of autophagy-related genes Beclinl in humen hepatocellular carcinoma, in order to explain that what is the function of Beclinl in HCC developing and it is significance.Method:We had collecte 43 cases of HCC tissues and 12 case of the normal tissue of hepatic angioma which had beed diagnosis by Dr. of the Department of Pathology of the Medical University of GuangXi from 2006,2-2006,9.We examed the expression of mRNA and protein of the Beclinl by RT-PCR and immunehistochemical (IHC).The result were used to analyse the relationship between the expression of Beclinl mRNA and protein in HCC and the information of pathology and clinic (including hepatitis B virus pathological stage,the level of serum AFP,the diameter of tumor and the presence of metastasis).Result:1.The RT-PCR consequence of the expression of Beclinl mRNA in HCC tissue,the HCC surrounding tissue and the tissue nearby hepatic angioma, the results respectively are 0.52±0.35,0.50±0.32,0.27±0.10.The difference between HCC tissue and the HCC surrounding tissue is not remarkable(P> 0.05),but HCC tissue and the HCC surrounding tissue compared with the tissue nearby hepatic angioma,the difference is remarkable (P<0.05).The position rate of Beclinl mRNA in HCC was significantly correlated with HBV(P<0.05),but not correlated with age,pathological stage,the level of serum AFP,the diameter of tumor and the presence of metastasis(P>0.05).2.The IHC consequence of the expression of Beclinl protein in HCC,HCC tissue,the HCC surrounding tissue and the tissue nearby hepatic angioma,the results respectively are 41.9%,51.2%,8.3%.The difference between HCC tissue and the HCC surrounding tissue is not remarkable(P>0.05),but HCC tissue and the HCC surrounding tissue compared with the tissue nearby hepatic angioma,the difference is remarkable (P< 0.05).The position rate of Beclinl protein in HCC was significantly correlated with HBV(P< 0.05),but not correlated with age,pathological stage,the level of serum AFP,the diameter of tumor and the presence of metastasis(P>0.05).Conclusion:①The expression of Beclinl between HCC tissue and the HCC surrounding tissue is not remarkable,but the expression of Beclinl in HCC tissue and the HCC surrounding tissue are higher than in the tissue nearby hepatic angioma, we suppose that the high level of expression of Beclinl might be related to the HBV,the alteration of the tumor growing surrounding and the defensive reaction.②The expression of Beclinl in HCC was correlated with HBV(P<0.05),but not correlated with age,pathological stage,the level of serum AFP,the diameter of tumor and the presence of metastasis(P>0.05).
