Cytotoxicity Of Enflurance, Isoflurane, Sevoflurane And Desflurane On LO2 Cells Cultured In Vitro

Objective: To evaluate the effects of enflurane, isoflurane, sevoflurane and desflurane on proliferation , biochemical index and secretion of Albumin of LO2 cells cultured in vitro.Method: Hepatic cells LO2 were cultured in vitro. Each drug, including halothane, isoflurane, sevoflurane and desflurane ,was divided into A,B,C and D groups. Respectively, A,B,C and D represented the plasma concentration of each drug at 1,2,5,10MAC. MTT assay was conducted to detect the effects of inhaled anesthetic for 24h on the viability of hepatic cells. Morphological change of such treated LO2 hepatic cells was also observed by versatile inverted microscope and captured. Then the activities of ALT and AST in the cultured medium of hepatic LO2 cells treated with inhaled anesthetic for 24h were measured by Reitman-Frankel. Finally, cell medium Albumin is measured by ELISA.Results: MTT results showed that there were no significant inhibition of proliferation in LO2 cells when the dosage of isoflurane, sevoflurane, and desflurane at 1MAC, but other dose groups all could inhibit the growth of LO2 cells in some degree. Enzyme activity assay results showed that the four drugs in the dose of 1MAC didn't influence the activity of ALT and AST in LO2 cells. Even the dose up 2MAC, isoflurane and desflurane didn't significantly increase the activity of ALT and AST. However, ALT and AST were markedly elevated in other dose groups. Albumin measurement results showed that albumin content in LO2 cells had no significant change when the dose of the four drugs at 1MAC. Even the dose up 2MAC, desflurane did not affect the ability of LO2 cells to secrete albumin. The albumin levels were significantly reduced in remaining dose groups.Conclusion: Hepatotoxicity of the Enflurane, isoflurane, sevoflurane and desflurane on the LO2 cells were strengthened with the drug concentration increased. The hepatotoxicity order of the four drugs is as follows: enflurane>sevoflurane > isoflurane > desflurane. Inhibiting of liver cell proliferation, damaging the liver cell membrane and inhibiting liver cell secretion of albumin are important hepatotoxic manifestation of inhalational anesthetics.
