Objective: Lymphatic metastasis is the main metastasis pathway in carcinoma of colon,but the concrete mechanism still is not clear.The experiment is to use the immunohistochemical staining to study the distribution and morphological character of microlymphtics in normal colon tissues ,colon low-grade intraepithelial neoplasia, colon high-grade intraepithelial neoplasia,carcinoma of colon.To analysis the change in normal colon tissues, colon low-grade intraepithelial neoplasia, colon high-grade intraepithelial neoplasia,carcinoma of colon,in order to discuss the ralationship of the number of lymphangiogenesis, the early diagnosis of carcinoma of colon and the prong of carcinoma of colonMethod: Income group of standards selected by the first visit without any treatment of carcinoma of colon development and progression stages of colon single site of several major organizations paraffin block. 28 cases of normal colon tissue, 42 cases of colonic high-grade intraepithelial neoplasia,colon low-grade intraepithelial neoplasia, 42 cases of colonic high-grade intraepithelial neoplasia, 42 cases, 42 cases of carcinoma of colon, carcinoma of colon at the junction of 42 cases, 42 cases of carcinoma of colon side. With D2-40 immunohistochemistry staining, observed under an optical microscope distribution of lymphangiogenesis occurred, the number of statistical lymphangiogenesis, describing the development of new lymphatic vessels in carcinoma of colon of all stages of the process of distribution, combined with follow-up data of new lymphatic control the occurrence of carcinoma of colon, development and prognosis.A new stage of development of carcinoma of colon occurs in the morphological characteristics of lymphatic vesselsResults: 1 Morphologic characteristic of the new lymphatic vessels in the development stage of carcinoma of colon.1.1 Enzyme histochmistry staining for D2-40 in normal colonic mucosa: Lymphatic vessels were D2-40 positive(brown).In normal colonic mucosa, Lymphatic vessels were mainly distributed in the muscularis mucosa, submucosa, muscularis and subserosa. Lymhpatic vessels have a more irregular and larger lumen,and a thinner wall parallel.There were no obvious lymphatic vessels in the lamina propria.1.2 Enzyme histochmistry staining for D2-40 in low-grade intraepithelial neoplasia: Lymphatic vessels were D2-40 positive(brown).There are a large number of lymphatic vessels in middle of the glands in lamina propria. It looks like trabs,and was connected into net.1.3 Enzyme histochmistry staining for D2-40 in high-grade intraepithelial neoplasia: Lymphatic vessels were D2-40 positive(brown).There are a more number of lymphatic vessels in middle of the glands in lamina propria of high-grade intraepithelial neoplasia than low-grade intraepithelial neoplasia. It looks like trabs,and was connected into net.1.4 Enzyme histochmistry staining for D2-40 in carcinoma of colon: Lymphatic vessels were D2-40 positive(brown).There are a large number of lymphatic vessels in middle of the glands in lamina propria. It looks like trabs,and was connected into net. By side of tumor, morphologic characteristic of the lymphatic vessels in mucosa is same to the normal colonic mucosa. 2 Relationship between the quantity of lymphangiogenesis and carcinoma of colon2.1 Compared the number of new lymphatic vessels in the normal colonic mucosa, low-grade intraepithelial neoplasia, high grade intraepithelial neoplasia, carcinoma of colon:the number of new lymphatic vessels in the normal colonic mucosa, low-grade intraepithelial neoplasia, high grade intraepithelial neoplasia, carcinoma of colon were (7.93±2.36),( 33.14±11.96),( 65.40±23.13) and (42.20±17.20), the number of samples were compared between groups,P <0.05.2.2 Compared the number of new lymphatic vessels in carcinoma of colon nests,the junction in carcinoma of cancer, the side of carcinoma of colon: the number of new lymphatic vessels in carcinoma of colon nests,the junction in carcinoma of cancer, the side of carcinoma of colon were (37.59±17.20), (46.82±17.20) and (7.20±2.32) respectively, the number of samples were compared between groups , P <0.05.2.3 Compared the number of new lymphatic vessels in the death group, the metastasis group, the recurrence group, the survival group of carcinoma of colon:The number of new lymphatic vessels in the death group,the metastasis group, the recurrence group, the survival group of carcinoma of colon are (56.07±18.68), (44.28±15.55), (27.56±5.45) and (31.54±2.08) respectively, the number of new lymphatic vessels in the death group, the recurrence group, the survival group samples were compared, P <0.05.The number of new lymphatic vessels in the death group and the metastasis of groups samples were compared, P=0.568>0.05.Conclusion: 1. In the course of the development of carcinoma of colon, the number of new lymphatic vessels in high-grade intraepithelial neoplasia group, carcinoma of colon group, low-grade intraepithelial neoplasia, normal colon tissue showed a decreasing trend. 2. There are the increasing number of new lymphatic vessels in the stage of high-grade intraepithelial neoplasia , it suggests that lymphangiogenesis can be used as an early indicator of malignant transformation of adenoma. 3. The number of lymphangiogenesis of carcinoma of colon is significantly related to metastasis and prognosis in carcinoma of colon.If there are more numbers of new lymphatic vessels in carcinoma of colon, there are the higher risk of metastasis and death in carcinoma of colon. the number of new lymphatic vessels may serve as the prognosis indicators of carcinoma of colon in pathology.
