Objective Recombinant human endostatin (Endostar) is the specificity of vascular endothelial growth factor, a large number of studies suggest that it could inhibit tumor growth through the inhibition of tumor angiogenesis. In our country, stomach is a common gastrointestinal malignancy, but a lower incidence in the United States, so there are no related research reports on the application of endostar in the gastric cancer. Studies suggest that angiogenesis of gastric cancer more than the surrounding tissue, so endostar used as a single agent to treat gastric cancer may be efficient. The experiment examine the inhibition rate of cell growth by MTT and view the effect of the apoptosis rate and the Snail and E-cadherin protein expression when cells were treated with endostar. And to investigate the effects of endostar on proliferation and apoptosis of gastric cancer cell line to provide theoretical and experimental basis for the clinical application.Methods MTT assay was used to examine the inhibition rate of cell growth when cells were treated with various concentrations of endostar (50,100,200,400ug/ml) for 72 hours; the apoptosis rate was examined by flow cytometry; invasion ability was examined by Transwell invasion assay; and Snail, and E-cadherin expression were examined by Western blot.Results (1) Effect of endostar on the proliferation:The inhibition rate of growth was enhanced in a dose-dependent manner when NCI-N87 cells were treated with endostar, the inhibition rate increase significantly with the degree of concentration. The value of CI20 was 150ug/ml. (2) Effect of endostar on the apoptosis:When the cells were treated with 150ug/ml endostar, the apoptosis rate was 32.83%, and the cell cycle was arrested in the G1 phase(78.64%). The number of S phase cells was small(3.17%).(3) Transwell invasion assay:The number of NCI-N87 cells permeating septum of Matrigel was 215.65±4.041 when the cellswere treated with endostar, and the control group was was 121.33±5.859. The invasiveness of the cells were obviously inhibited by Endostar.(4) Effect of endostar on Snail, and E-cadherin expression:Compared with control group, when NCI-N87 cells were treated with endostar, Snail expression was strong positive, but E-cadherin expression was poor positive.Conclusion:The recombinant human endostatin(Endostar) can inbibit the growth and induce the apoptosis of NCI-N87 cells in vitro, and may lead to decreased expression of Snail and E-cadherin. Endostar has antagonism effects on the growth and invasion and metastasis and of gastric cancer cell NCI-N87.
