Also chosen closely related with the ischemia-reperfusion injury is cytokines IL-I, and rabbit renal pathology and electron microscope, at third days and seventh days, as evidence of clinical effectiveness was observed on rabbit renal damage, and to investigate the retrograde perfusion kidney to improve the function and structural damage in IRI, Improve the theoretical basis for clinical feasibility of retrograde perfusion donor kidney.Method:First To establish a rabbit kidneys model after retrograde perfusion of IRI,and the establishment of rabbit kidneys antegrade perfusion group and non-perfusion group of IRI in the two control groups.The 30 adult male New Zealand white rabbits were randomly divided into 3 groups of 10 each, each group were randomly divided into two groups. the first group, the blood of the first day was take,specimens were taken for routine pathological examination and electron microscope on the third day. The second group of rabbit, respectively, also take the first day, second and third, the fifth day, seventh day the venous blood, the upper serum after centrifugation to SCr and BUN were measured, specimens were taken for routine pathological examination and electron microscopy on the seventh day. At last,Determinate the rabbit IL-1 concentration about the first day of each group.Result:The serum Cr and serum BUN of the antegrade perfusion groups increased fastest,There were significantly different compared with that of the retrograde perfusion group and the non-perfusion group (P<0.01), with the latter two were no significant differences compared (P>0.05). The serum Cr, serum BUN of three groups rabbit returned to normal in the fifth day.There was a positive correlation between levels of routine pathological examination of kidney and increase of The serum Cr and serum BUN.and electron microscope so. The IL-1 concentration of the antegrade perfusion group is highest in the first day,then that of not perfusion group is second, and that of the retrograde perfusion group is minimum, That of the antegrade perfusion group was statistically significant (P<0.01)than that of the retrograde perfusion group and that of not perfusion group, That of the retrograde perfusion group were not significantly different (P>0.05) than that of the non-reperfusion group.Conclusion:1. Establishment of a rabbit kindneys model of retrograde perfusion on renal cold ischemia reperfusion injury in situ were more successful, feasible, which can meet the needs of experimental study.2.The retrograde perfusion kidney in cold ischemia-reperfusion injury to improve the function and structural damage with the conventional renal perfusion due to ischemia-reperfusion injury and non-perfusion ischemic reperfusion injury group have better effect.3. Reverse perfusion may bring more safe,convenient means for renal transplantation. |
