| Acute coronary syndrome(ACS)is a common illness to harm seriously to the people's healthy,including unstable angina pectoris(UAP),ST-segment elevation myocardial infarction(STEMI) and non.ST-segment elevation myocardial infarction (NSTEMI).ACS occurs when unstable atherosclerotic plaque ruptures,ulcerates and blood platelet assembling to follow thrombus formation,which results in sudden total or nearly total arterial occlusion and myocardial infarction.ACS is a complex multifactorial disorder which is believed to result from the interplay between a person's susceptible genetic makeup and various environmental intricate factors.The interaction of multitude predisposing is pathopoiesis foundation of ACS,these interaction emerge phlegmonosis stimulating factor,which leads to rupture of instability plaque in coronary artery and forms ACS.Pregnancy-associated plasma protein-A(PAPP-A) is a inflamming correlation factor,which is a researching warm spots recently and expresses abundantly into instable atheromatous plaques,it is a sensible instability plaque marker.Domestic and oversea reaseach shew that the blood serum level of PAPP-A was a aid index as diagnostic ACS and was significant value.in identificating hazard layer and judging prognosis.The reaseach oversea shew that there was significantly association between the IVS 6+95C allele of PAPP-A gene and AMI.These articles have not been reported whether the gene was relationship with ACS among the Chinese Han population.This article explored the frequences of genotype and allele distribution and the possible relationship of PAPP-A IVS6+95 gene Polymorphism with ACS in the Chinese Han population of Sunan region.Objective:To investigate the distribution of PAPP-A gene IVS6+95 polymorphism in the intron region and the association of ACS in the Chinese Han population of Sunan region. Methods:424 patients (ACS group) and 516 control subjects (control group) who were free from coronary artery disease confirmed by cardioangiography were recruited to participate in this study,these patients included 172 UAP and 252 AMI.The genotype of PAPP-A IVS 6+95 polymorphism was determined by polymerase chain reaction and restriction fragment length polymorphism analysis. And at the same time, we measured the plasma levels of blood sugar,total cholesterol(TC),triglyeride(TG),low density lipoprotein-cholesterol(LDL),high density lipoprotein-cholesterol(HDL) and so on in WuJin People's hospital. SPSS13.0 software package was applied to carry out statistical analysis.Results: There were CC CG and GG genotypes of PAPP-A gene IVS6+95 polymorphism in the ACS group and control group,the two groups could be considered as a genetic equilibrium representative by Hardy-Weinberg equilibrium (P>0.05).As compared with the control group, the frequencies of CC,CG,GG genotype and C allele (8.25% vs 5.43%,48.35% VS 43.02%, 43.40% vs 51.55% and 32.20% vs 26.94%, respectively) were not significantly different in ACS group;But multivariate logistic regression analysis adjusting for traditional ACS risk factors such as smoking,blood pressure,blood sugar,TG,TC,LDL,HDL indicated that there was significant correlation between the CC genotype,C allele and ACS (OR=0.948,95% CI:0.709~1.267,P=0.044;OR=0.948,95%C/:0.709~1.267,P=0.049 respectively). When the female were carried out statistical analysis,there was close correlation between PAPP-A IVS6+95CC genotype and ACS (p=0.001). There was significant difference between the AMI group,the UAP group with and normal coronary group.Conclusion:1 .The crowd existed the IVS 6+95 polymorphism of PAPP-A gene in the Chinese Sunan region2.There was significantly association between the IVS 6+95 single nucleotide polymorphism (SNP) of PAPP-A gene and ACS.The present findings suggested that the CC genotype of PAPP-A gene IVS6+95 polymorphism may be associated with an increased risk for ACS,the C allele may be responsible for the patient of ACS in the region especially to female and the SNP is associated with the risk of AMI and UAP among the Chinese Han population of Sunan region. |
