Ramulus mori is the rhizome of Morus alba L.The article is about the water-soluble polysaccharide from ramulus mori,The composition and characterization of Ramulus Mori polysaccharide were researched by TLC,IR,GC,HPLC,smith degradation and so on.These work could establish foundation for the further study of Ramulus mori polysaccharides.1.With single factor and response surface methodology,the extraction technology of Ramulus mori polysaccharide is studied,The optimal extraction technology was was that the polysaccharides were extracted with water 20 times, 100℃,three hoursand twice extraction.The determination of Ramulusmori polysaccharide was 17.28%by phenol-sulfuric acid method.This method is convenient,and the stability and reproducibility are good.2.The protein in the Ramulus mori polysaccharides was removed by TCA and was further purified by DEAE ion exchange cellulose(DEAE-52)and SephadexG-100,received RMPs-1 and RMPs-2,They are single symmetrical peaks detected by HPLC.It concluded that the two amulus Mori polysaccharides are pure polysaccharide,the molecular weighe of RMPS-1 is 5.8×10~5 Da,RMPS-2 is 6.5×105 Da.3.The monosaccharide composition of RMPs-1 and RMPs-2 were studied by TLC and GC,RMPs-1was made up of rhammose,arabinose,glucose and galactose with the molarity rate of 1.08:1:1.40:1.57:3.80,RMPs-2 was made up of rhammose,glucose and galactose with the molarity rate of 24.0:1:1.34:12.61.4.The characterization of Ramulus mori polysaccharide was researched by IR,GC,HPLC,smith degradation and so on..The results were that Ramulus mori polysaccharide was brownish red colored powder,tasteless,water-soluble and undissolved alcohol,acetone,chloroform,et al.The content of arabic caid is 25%in RMPs-1,and 34.7%in RMPs-2.The UV spectrums and Infrared spectrum showed that they have the polysaccharide characteristic absorb peaks.Smith degradation showed that the main linkage form in RMPs-1and RMPs-2 was 1→2 and 1→4 glycosidic linkages,But some 1→3 glycosidic linkages also existed in the molecules; The experiment of congo red show that RMPs-land RMPs-2 have the spiral structure.5.When the concentration is small,RMPs-1and RMPs-2 promote the growth of the Hela tumor cells,And with the concentration increase,RMPs-1and RMPs-2 have the inhibition to the tumor cells,when the concentration of RMPS-1 increase to 500μg/mL,The inhibition rate reach to 24.98%and 29.93%.
