Objective:To study the expression of Toll-like receptor(TLR)2,TLR4 in the lesions and Peripheral Blood Mononuclear Cells(PBMC) of patients with Condyloma Acuminatum(CA) and the possible role of the two receptors in the pathogenesis of CA.Methods:The expression of TLR2 and TLR4 was detected in 34 specimens from lesions of CA and 20 from normal skin using the method of immunochemistry.Meantime,TLR2 and TLR4 mRNA were detected in the PBMC of the same patients by SYBER GreenⅠReal-time PCR,and those in health people(n=30) were detected as comparison.Results:Both TLR2 and TLR4 were expressed in the lesions of CA and most of them were strongly expressed.TLR2 was expressed in the prickle cell layer,and TLR4 was expressed in the entire epidermis. However,in the normal epidermis,TLR2 was weakly expressed in the basal cell layer and prickle cell layer in most of the specimens and the expression of TLR4 was mainly negative.Both the expression of TLR2 and TLR4 in the lesions of CA is higher than that in the normal skin(P<0.01).Whereas,compared with healthy people,the expression of TLR2 mRNA in patients with CA is not changed dramatically(P>0.05),and that of TLR4 mRNA is lower,but there is no statistical difference(P>0.05).Conclusion:The result that the expression of TLR2,4 in the lesions of CA is higher than that in the normal skin indicates that TLR2,4 can recognize HPV,they relate to the pathogenesis of CA and they may play an important role in the immune process of virus infection.And compared with healthy people,that the expression of TLR2,4 mRNA in the PBMC of patients with CA does not increase may be the result of the incompetence of HPV invading into the blood and may relate to the immune restraint of patients with CA.
