Objective In this study, we established animal models of rabbits with acute myocardial ischemia-reperfusion injury, to observe the effect of roxithromycin intervention on rabbits with acute myocardial ischemia-reperfusion injury and observing the change of inflammatory cytokines and Europhile apoptosis. to research possible mechanisms Roxithromycin played, to provide the basis for clinical prevention and intervention about acute myocardial ischemia-reperfusion injury.Methods Thirty healthy male New Zealand white rabbits, general food to feed a week, were randomly divided into 3 groups (n=10 rats):sham operation group (Sham group) only anterior descending coronary artery ligation no threading; ischemia-reperfusion group (IR group) left anterior descending coronary artery ligation threading 30 minutes, then release the thread and reperfusion for 180 minutes; Roxithromycin intervention group (Apo group) to give oral roxithromycin 10mg/kg, day 1, used in conjunction three days after the preparation of animal model of ischemia-reperfusion., Respectively, before ischemia,30 minutes after ischemia and reperfusion 180 minutes after blood collection using thiobarbituric acid (TBA method) plasma malondialdehyde (MDA) content; end of the experiment, the animals were killed quickly, with Evans Blue-TTC double staining, myocardial infarct size, myocardial specimens were obtained morphology (HE staining, transmission electron microscopy), detection of peripheral blood interleukin-6,8,10 (IL-6, IL-8, IL-10) and tumor necrosis factor-α(TNF-α) levels and neutrophils (PMNs) apoptosis. The experimental data was analyzed by using SPSS 11.0 software. Multiple comparison use single-factor analysis of variance, pairwise comparison between the two groups use q test, time-dependent variable data use of repeated measure analysis of variance of data.Results1.Sham group, IR group and Apo group before ischemia MDA showed no significant difference.2. Ischemia Apo group and the IR group than in Sham group MDA slightly higher (P<0.01) but the IR group compared with the Apo group no significant difference between MDA, suggesting that the two groups to establish a successful model of ischemia and no statistically significant difference.3. Reperfusion after the end of Apo group and IR group than in Sham group of MDA increased (P<0.01) and significantly higher in IR group(P<0.01), prompted the establishment of ischemia-reperfusion model of success, and the roxithromycin group rabbits with acute myocardial ischemia-reperfusion injury in a lighter.4. Reperfusion group after the end of Apo compared with the IR group IL-6, IL-8, TNF-a content decreased, IL-10 levels increased, rise in the rate of neutrophil apoptosis, myocardial infarct size decreased (P<0.01), optical lens shape to improve the myocardial injury, suggesting that roxithromycin can reduce the inflammation factor, increase the inflammatory response to a negative regulatory factors, induced neutrophil apoptosis and improve myocardial ischemia-reperfusion injury.Conclusion Roxithromycin pretreatment on rabbits with acute myocardial ischemia-reperfusion injury in a significant protective effect, this protective effect may be related to reduced cardiac inflammatory cytokines, increase inflammation in the negative regulatory factors, induced neutrophil apoptosis, thereby reducing activity of oxygen damage, anti-oxidation to enhance and protect endothelial normal function, reducing neutrophil-related myocardial damage.
