Background:Since the concept of MIRI is firstly be issued by Hearse in 1977, prevention and treatment of MIRI have been focus of research by many specialists with development on diagnosis and treatment of acute coronary syndrome including coronary artery bypass grafting, percataneous coronary angioplasty and so on. Reasonable selection of drugs is a common and effective method on myocardium protection antagonizing MIRI. Flavonihippophae is a kind of flavonoids taken from scarce and precious Fructus Hippophae with 40 million years history, which is verified by experiment and clinic to have functions such as anti myocardial ischemia, decrease of myocardium oxygen consumption and increase of myocardium function. However, the there's no reports about its protection and mechanism on ischemia reperfusion myocardium. Losartan belongs to angiotension II receptor blocker which could improve myocardium ischemia, decrease infarction area and obviously protect MIRI injury verified by experiment and clinic. The research is to observe the change of MMP-9,TIMP-1,MMP-9/TIMP-1; Bcl-2,Bax,Bcl-2/Bax in ischemia-reperfusion myocardium tissue in comparison with losartan, to explore protection of flavonihippophae on MIR injury by combing with change of ultra structure and to further study on its mechanisms. It will provide experimental data for clinic prevention of MIRI and seek for an effective and natural drug protecting reperfusion myocardium in order to decrease long term mortality of acute myocardial infarction patients, increase live quality of patients and give impetus to rapid development and dispersion of intervention therapy of coronary heart disease.Objective To observe the effects of The Flavonihippophae on Myocardial Ischemia-Reperfusion Injury and the MMP-9,TIMP-1,MMP-9/TIMP-1 and Bcl Family (Bcl-2,Bax,Bax/Bcl-2) expression in cardiac tissues of rats, and to explore theprotective mechanism of The Flavonihippophae on Myocardial Ischemia Reperfusion Injury.Methods 50 rats were classified 4 groups by using the random grouping principle:model group, losartan group, Flavonihippophae group by using different dose and sham operation group. the Myocardial Ischemia-Reperfusion Injury model was established by ligating left anterior descending coronary artery for 30 minutes and releasing then. The cardiac muscle tissue was stained by HE to observe its necrosis area and pathological changes as well as the expression of MMP-9,TIMP-1,MMP-9/TIMP-land Bcl Family (Bcl-2,Bax,Bax/Bcl-2) by immunohistochemistry method and Western-blot.Results Method HE sections show that Necrosis of cardiac muscle in rats is significantly relieved in Flavonihippophae group by using different dose compared with model group,The immunohistochemistry and Western-blot method result show that①The Flavonihippophae group using different dose decreased the expression of MMP-9 and MMP-9/TIMP-1 compared with the model group and increase the expression of TIMP-1 (P<0.05);②The Flavonihippophae group using different dose decreased the expression of Bcl-2compared with the model group and increase the expression of Bax and Bax/Bcl-2 (P<0.05).Conclusion Flavonihippophae has protective effect on cardiac muscle after the Myocardial Ischemia-Reperfusion Injury, and its mechanism may be related to:①Inhibition of MMP-9 and MMP-9/TIMP-1 and raise TIMP-1 expression in cardiac muscle;②Inhibition of decrease Bax and raise Bcl-2andBcl-2/Bax expression in cardiac muscle.
