| Objective:Melamine and cyanuric acid were administered by oral gavage to establish the animal model of kidney injury and calculus induced by the melamine and its analogues (cyanuric acid). For further research of an experimental basis, pathological characteristics and stone formation are investigated.Methods:female SD rats were divided into control group,melamine group, melamine-cyanuric acid group, according to the daily per kilogram of body weight amount of melamine and cyanuric acid, were randomly divided into A, B, C three groups.The rats have been administrated by gavage for 1 week. Fed rats were detected serum creatinine, blood urea nitrogen when rats were administrated for one week and at 2 weeks of administration ceased,all rats were euthanized by ether to observed renal pathological change.Results:1.At 1 week after administration, the body weight of control group and melamine group increased, on the other hand the body weight of C and B group decreased significantly,and the fur of some rats become withered and yellow.2.After administration, compared with the control, serum creatinine and blood urea nitrogen level in B group and C group increased.there were no statistical differences in A group and melamine group.At 2 weeks after administration ceased, serum creatinine and blood urea nitrogen of the rats were different from the rats administrated after one week in B group and C group.3. After administration, the volume of kidney swelled in all treated rats. In the B group and C group, the color of kidney become light yellow,and the corticomedullary junctions were obscured.In the C group,3 rats died after administration.On section,there were not abnormal in control group,and glomerular infiltration of lymphocytes were only seen in melamine group. Renal tubular edema, epithelial shedding and the formation of stone crystals were observed in some rats of A group.In B and C group, renal tubular edema, epithelial shedding were more severe in all rats, some of which appear glomerular fibrosis, and crystals have been observed in all histological sections.Conclusion:1. Melamine have just been administrated for 1 week, there are no obvious pathological kidney lesions and crystal stones.2. Melamine and cyanuric acid were administrated simultaneously, the volume of kidney and weight increased, and pathological lesions present in glomerular and renal tubule. The extent of pathological lesions and the amount of crystal formation increased depended on the level of melamine and cyanuric acid.3. A kidney lesions and kidney calculus experimental model induced by melamine and cyanuric acid will be established by administrating melamine and cyanuric for animals. |
