ObjectiveBased on the alcoholic liver disease rat model,we investigated the major pathological mechanisms of experimental alcoholic liver disease .To explore the effects and mechanisms of betaine preventing alcoholic liver disease.To observe the relationship of plasma total homocysteine (tHcy) level and plasma betaine level in rats.Method35 male Wistar rats and 35 female Wistar rats (the average weight was 200±20g)were randomly divided into five groups(each group with 7 male rats and 7 female rats)according to their weight.The five groups were normal-group ,alcoholic model group ,betaine in low-dose group,betaine in middling dose group and betaine in high dose group.In the alcoholic model group,wister rats were treated with 40% (V/V) ethanol 8g/Kg intragastric infusion twice a day to the end of the forth week,then,from the beginning of the fifth week,treated with 50%ethanol(V/V)9g/Kg.d intragastric infusion twice a day to the end of the eighth week.Rats in the betaine preventive group were fed different-dose betaine (100mg/kg,200mg/kg and 400mg/kg) to intervene.Rats in the normal groups were fed 0.9% NaCl.At the end of experiment, animals were sacrificed, the fasting liver and blood samples were collected .Selected samples of the left liver from each rat were rapidly removed at random and fixed in 10% neutral buffered formalin.The sections were stained with hematoxylin and eosin(HE)and oil red.,and the histopathological analysis were made.The rat serum ALT,AST,TG,TC,TNF-αlevels were evaluated using assay kits. Selected samples of right liver from each rat were rapidly stored under -80℃for use.The protein content was estimated by the dye-KaoMaShi,SOD,GSH,MDA in liver tissue were evaluated according to the manual of assay kit.Plasma levels of Hcy and betaine were respectively measured by ELISA and HPLC . All date were expressed as x±S, and the analysis were performed with SPSS12.0. Statistical significance was assigned at a P value of <0.05 or<0.01.Results1.Light microscopic observed the histology of the liver with HE or oil red dying :the typical injury change of chronic alcoholism were observed in rats of alcoholic model group.The injury of liver was alleviated in rats of different dose of betaine.The rats of normal group showed the normal histology of liver.2.The AST,ALT,TG,TC,TNF-αlevel in serum of alcoholic model group were significantly increased compared with the normal group.The AST,ALT,TG,TC,TNF-αlevel of the groups of different dose of betaine were significantly decreased compared with the alcohol model group.3.The activity of SOD and the content of GSH in the liver tissue of the alcoholic model group was significant lower and the content of MDA was higher compared with normal group. The activity of SOD of the groups of different dose of betaine were significant higher compared with the alcoholic model group .The content of MDA of the groups of different dose of betaine were evident lower than that of the alcoholic group.The content of GSH of the groups of different dose of betaine were higher compared with the alcoholic group,especially in the middle and high dose 's group of betaine.4.The plasma tHcy of the significant higher compared with the normal group.The plasma Hcy of the groups of different dose of betaine were decreased compared with the alcoholic model group.Plasma tHcy level and plasma betaine level were inversely related (r=-0.448,P<0.01)in rats.Conclusion1. Betaine might protect the liver function in rats of alcoholic liver disease.Betaine could decrease the level of serum AST and ALT and ameliorate hepatic pathology in rats of alcoholic disease.2..Betaine could decrease the level of plasma tHcy in rats of alcoholic disease.Plasma tHcy level and plasma betaine level were inversely related in rats.3.Betaine could decrease the level of serum TG and TC in rats of alcoholic disease. 4.Betaine could increase the antioxidant ability in rats of alcoholic liver disease .5. Betaine restrain the secretion of the TNF-αand decrease hepatic necroinflammatory.
