Objective To investigate the clinical significance of the expressions of survivin gene and its main transcript variants mRNA in colorectal carcinoma.Materials and Methods The mRNA expressions of total survivin, survivin-△Ex3, survivin-2B,survivin-3B, survivin 2αand CEA CAM 5 in tumor tissues, adjacent tissues (2.5cm beyond the cancer margin) and norma1 colorecta1 tissue (10cm beyond the cancer margin) from 39 cases of colorecta1 cancer were detected by RT–PCR. Total mRNA levels of survivin and survivin-△Ex3 were scanned and quantified with gel imaging analytical system, and of survivin-2B, survivin-3B, survivin 2αand CEA CAM5, by SYBR GreenⅠreal-time fluorescent semi-quantitative PCR.Results①The positive rates of total survivin, survivin-△Ex3, survivin-2B, survivin-3B, survivin 2αand CEA CAM5 mRNA in tumor tissues (100.0%, 92.3%, 92.3%, 79.5%, 87.2% and 97.4%) were significantly higer than those (38.5%, 25.6%, 28.2%, 7.7%, 12.8%, and 12.8%) of adjacent tissues (P<0.01), and those (17.9%, 5.1%, 7.7%, 0.0%, 2.6% and 10.3%) of norma1 colorecta1 tissues (P<0.05).②In all samples of positive expressions, the total mRNA levels of survivin, four transcript variants and CEA CAM5 were significantly higher in tumor tissue (2.36±1.41), (0.76±0.35), (0.49±0.27), (0.08±0.03), (0.07±0.03) and (23.72±10.38) than those of (0.88±0.54), (0.32±0.17), (0.09±0.05), (0.02±0.01), (0.01±0.01) and (1.05±0.67) in adjacent tissues (P<0.01), respectively. However no significant diference was found between adjacent tissues and norma1 colorecta1 tissues (only total surviving (0.57±0.11), survivin-2B (0.05±0.02) and CEA CAM5 mRNA (0.77±0.45) have data to compare) (P>0.05).③Several transcript variants mRNA levels were associated with the clinicopathologic parameters of patients, The expressed levels of survivin-△Ex3, survivin-3B and CEA CAM5 mRNA were associated with Dukes grade and lymphoid metastasis (P<0.05); Survivin-△Ex3 and CEA CAM5 mRNA were associated with invasive depth and pathological grade of the patients, respectively.Conclusions①Measurements of survivin transcript variants mRNA expressions may be more useful than CEA mRNA in the diagnosis and prognosis of colorectal carcinoma.②The mRNA levels of surviving and its transcript variants were highly expressed in colorectal carcinomas, several transcript variants mRNA levels were associated with clinicopathologic parameters of malignancy.
