Objective To detect proportion of CD3~+ T-lymphocyte in peripheral blood mononuclearcell and CD3~+/CD30~+,CD3~+/CD154~+ T-lymphocyte in CD3~+ T-lymphocyte cluster,peripheral blood platelets count,and apoptosis rate of activated T-lymphocyte in chronicity ITP patients,approach the immunity disord state and the effect of these signaling molecule in this disease.To build up information for research this disease,expect to search suitable target for immunologic intervention treatment.Methods random choice 40 cases of chronicity ITP patients for investigate group and 24 cases of health people for control group,sterility collecte peripheral venous blood,measured proportion of CD3~+ T-lymphocyte in peripheral blood mononuclearcell,CD3~+/CD30~+,CD3~+/CD154~+ T-lymphocyte in CD3~+ T-lymphocyte cluster and actived T-lymphocyte Cell apoptosis rate in mononuclearcell of peripheral blood by flow cytometry measuremen,simultaneously measured peripheral blood platelets count. To analyze difference and correlation between these targets.Results①The rate of CD3~+ T cell have no difference in ITP and healthy control group(51.74±12.86 vs 49.81±10.79,P>0.05).②The activated T-lymphocyte apoptosis rate in ITP was lower than healthy control (27.64±11.65 vs 45.71±12.37,P<0.01).③The apoptosis rate of activated T-lymphocyte was positive correlation with platelets count(r=0.680,P<0.01).④The CD3~+/CD154~+ T cell in ITP was lower than healthy control(2.85%±2.22%vs 6.17%±3.01%,P<0.01) and positive correlation with it's apoptosis rate(r=0.462,P<0.05).⑤The CD3~+/CD30~+ T cell in ITP were higher than healthy control(9.39%±2.58%vs 2.82%±0.49%,P<0.01) and negative correlation with it's apoptosis rate(r=-0.786,P<0.01).⑥The expression of CD30 in chronicity ITP were negative correlation with CD154(r=-0.593,P<0.05).Conclusion 1. The chronicity ITP patients' actived T-lymphocyte Cell living time prolonged and apoptosis inhibited.Indicated there were activation T-lymphocyte Cell accumulated,that maybe an important reason for auto-chthonous immune function confused.2.The CD154 depress expression and CD30 high expression on actived T-lymphocyte Cell is an important reason for AICD function confused in chronicity ITP patients.3.The CD154 and CD30 expression was intimate correlated,high expression of CD30 maybe an important reason of CD154 depress expression.4.According to investigate before,we suppose CD30 high expression may induced T-lymphocyte subgroup disturbance,secreted great quantity of Th2 cytokines, kill and wound platelet through hypersensitivity reactionⅡand mediate cytotoxic function. 5.The chronicity ITP patients' CD30 high expression on actived T-lymphocyte Cell can be a target for treat.Apoptosis depress intimate correlated with peripheral blood platelets count, can be a index for pathogenetic condition and curative effect monitoring.
