Objective:The diagnosis of osteoarthritis(OA) is based upon the clinical examination and the radiographic assessment,both of which can be non-specific and insensitive in the early diagnosis of OA.Biochemical indicators in synovial fluid can reflect changes of joint tissue quantitatively and dynamicly,so it can be an important index helping to diagnose OA.The purpose of this study is to detect the contents of IL-1βand IL-6 in synovial fluid of osteoarthritis patients,and discuss the relationship between the contents and the severity of knee osteoarthritis which reflected by arthroscope,then look for the biochemical indicators which can reflect the severity of osteoarthritis.Method:40 patients to accept knee joint arthroscope treatment or prosthetic replacement for knee joint were selected.11 patients with merely meniscus injury were divided into control group.29 patients with knee OA were divided into 3 groups according the severity of knee osteoarthritis which reflected by arthroscope.Synovial fluid was extracted under aseptic condition and be measured.Results:The concentrations of IL-1βbetween control group and experimental group have significantly different(P<0.01),the experimental group among the different classifications also significant(P<0.01).As to IL-6 test results,the control group and experimental groups don't have significant differences(P=0.09),but we think this may de caused by less sample size. Pairwise comparison found that concentrations of IL-6 in mild and moderate groups were significantly higher than control group and severe group(P<0.01),but the differences between the control group and sever group,mild group and moderate group don't have statistical significance.The concentrations of IL-1βin knee synovial fluid of OA patients and the severity reflected by arthroscope were positively correlated(r=0.887,P<0.01),and the concentration of IL-6 and the severity of OA is not a straight line relationship.Conclusion:The concentration of IL-1βwhich consists in the knee joint fluid of OA patients is obviously higher than healthy person,so it may be used as the biochemical indicator to distinguish the severity of OA,also can be the reference index to estimate the effect of taditional Chinese medicine in the treatment of OA.However,the concentration of IL-6 in the knee joint fluid of OA patients raises obviously in the early phase,and has a descendent trend in the later period,so it can not be the biochemical indicator to distinguish the severity of OA.
