Composed of two parts which include theoretical and experimental research, it discus about the Effect and its Mechanisms of YiFeiFang on inhibiting Proliferation of the human lung cancer cell A549.Theoretical study involve the clinical reach of Western medicine and Chinese medicine on the primary bronchogenic carcinoma, non small cell lung cancer especially during the past 10 years. Discussing the evidence of the benefit Qi and Yi ,also antidotal method treat the lung cancer from the characteristics of the lung's physiology pathological mechanism ,the causation of happen, clinical manifestation and so on.To investigate the effects of ganister on the proliferation and molecul mechanism on A549 cells by experiment of cells and molecule. A549 cells were treated with different does of YiFeiFang, the growth inhibition rates and cytotoxic effect of A549 cells were measured with MTT assay. Morpholiogical changes of the apoptosis of cancer cells were observed by AO/EB .To detect Yifeifang's effect on different gene expression of human lung cancer A549 cells tumor-related biological pathway by PCR gene microarray, analyse the characteristic biological pathway of its inducing gene spectrum changing, and discuss its anti-tumor molecular mechanism. through validation on the characteristics of the biological pathway on the regulation of gene expression by qPCR, to explore its anti-tumor effects of the molecular mechanism. The result of those experiments shows that , The inhibition of YiFeiFang toA549 cells was concentration dependent. The value of the IC50 for 48 hours was 5.75 mg/ml. Apoptosis in the experimtal group was observed by AO/EB. Detecting differentially expressed genes by PCR gene microarray shows that:8 of the 89 genes were detected up-regulated expression, and 22 were down-regulated expression in experimental group in comparition with the control group. The differentially expression was related to angiogenesis, apoptosis, cell senescence, invasion and metastasis and signal transduction. Differentially expressed genes of tumor-related biological pathway of A549 cells acted by Yifeifang were selected successfully by the technology of PCR gene microarray. It reflects that Yifeifang inhibits human lung cancer cells growth by the way of regulating gene expression of tumor-related biological pathway of angiogenesis, apoptosis, cell senescence, invasion and metastasis and signal transduction. Compared with control group, the concentration of different drugs in the experimental group of A549 cells VEGF, IL8, COX-2 expression levels were reduced to varying degrees by qPCR, and showed a certain degree of dose-effect relationship, of which COX-2 and with the administration of lower concentrations, there are upward trends, and IFNA gene 5mg/ml concentration of the drug under the conditions of the expression level increased significantly, and the remaining two groups are significantly lower expression levels. YiFeiFang on angiogenesis-related gene expression levels of the prominent role of regulation and control may be one of the molecular mechanism of the treatment of non-small cell lung cancer.
