Objective:To study the selective angiography of the vertebral-basal artery in cerebral vasospasm animal model after subarachnoid hemorrhage in rabbit via the transfemoral route;To study the Effect of transcranial Doppler Ultrasonography (TCD) evaluating cerebral vasospasm after subarachnoid hemorrhage in rabbits;To approach phosphodiesteraseⅤvariance in content and activity of CVS rabbits'basal artery in different period and to evaluate the correlation between phosphodiesteraseⅤand CVS.Methods:Vasospasm was induced in rabbits by the double injections of blood into the cisterna magna.To assess the effect of cerebral vasospasm and the blood flow rate of basal artery,the selective angiographies of the vertebral-basal artery and TCD were made before operation and the 1st,3rd,5th,7th,9th,11thday after operation successively.After TCD and DSA,the rabbits were sacrificed by cardiac perfusion mothod,and then we taken the basal arteries after digging skull operation. The artery specimen for routine histopathological and Immunohistochemical examination were put into Formalin for 1 week and then made paraffin-embedded sections for test.Other specimen were preserved in liquid nitrogen immediately after getting them.When the arteries were harden,we saved them in super low temperature refrigerator.The PDEⅤcontent variation in rabbits'basal arteries were detected by immunohistochemistry and Real-Time RT-PCR,and the activity changes were tested by high performance liquid chromatography.Results:The selective angiography of the vertebral-basal artery in cerebral vasospasm animal model after subarachnoid hemorrhage in rabbit was made successfully.Diameter of the basilar artery was assessed by angiography.The high quality and intensity of basilar arterial TCD image can be acquired by improving frequency of ultrasound signal,so that we can assess the variance of blood flow rate of the basal artery.The average vessel diameter of basal arteries in normal rabbits is 0.82±0.08mm,and the mean velocity of basal arteries in normal rabbits is 27.39±6.25 cm/s.The basal arteries diameter became thinner and the development time of arteries prolonged,and the mean velocity of basal arteries got fast in SAH group 24h after models finished.Which became most seriously 3 days after operation,the phenomenon last until 7 days after operation and then became better. All of them didn't became normal until lid after modered.The PDEⅤof basal artery in SAH began to express significantly 24h after operation.Its content increased and activity enhanced,which became most seriously 3 dys after operation,the phenomenon last until 7 days after operation and then became better. All of them didn't became normal until lid after modered.The diameter and velocity of basal arteries didn't changed significantly in blank control group and experimental contrast group,and the PDEⅤcontent and activity didn't changed too, which didn't express obviously in these two groups.Conclusion:This experiment confirm that it is feasible to make the selective angiography of the vertebral-basal artery in cerebral vasospasm animal model after subarachnoid hemorrhage in rabbit via the transfemoral route.It is easy and accurate to assess the blood flow rate of basal artery successively by TCD.There is good dependability between TCD and DSA in assess the cerebral vasospasm after subarachnoid hemorrhage in rabbit.The PDEⅤcontent of basal artery in SAH rabbit increase and the activity enhanced,which is closely related to the test of CVS by TCD and DSA in time.So the PDEⅤmay be related to the CVS after SAH.
