Introduction: Lymphangiogenesis was thought to be involved in many pathophysiological process such as tumor wounds and inflammation. It was believed in the past investigation that :there were two major routes in the lymphangiogenesis, the new born tlymphatic vessals were budding ftom he intrinsical lymphatic vessals, and the endothelial progenitor cells in peripheral blood could be induced by VEGF-C to differentiated into lympahtic endothelials. Monocytes migrate from blood circulation to local tissue transformate into macrophages in the pathophysiological process as tumor wounds and inflammation, produced VEGF-C, palying an important role in the lymphangiogenesis. It has been discovered in the present study that monocytes could be effected by VEGF and son on to transdifferentiate into blood vessal endothelials,however,there were no denmonstration whether the monocytes could be induced into lymphatic endothelials in vitro. Our research focused on the monocytes stimulated by fibronectin, lipopolysaccharide, TNF-α,VEGF-C, IL-3,IL-7 which were produced by the tissue in inflamational conditions.and the markers of lymphatic endothelials LYVE-1,Podoplanin,Prox1 and endothelials markers vWF, eNOS were studied, meanwhile the possibility of the transdifferentiation of mononuclear-macrophages into lymphatic endothelial cells in inflammatory condition was studied. and could provide theorial evidence for the research of lymphangiogenesis.Objective: Lymphangiogenesis is thought to be involved in the pathophysiological process such as tumor and inflammation. Here, the possibility of the transdifferentiation of mononuclear-macrophages into lymphatic endothelial cells in inflammatory condition was studied.Results: Before the induction, the expression of LYVE-1 was detected positive in cultured mononuclear-macrophages, while the expression of vWF, eNOS, Podoplanin, Prox1 was detectednegative. After the induction, the level of expression of Podoplanin and Prox-1 obviouslyincreased in cultured mononuclear-macrophages, yet the expression of vWF and eNOS remainednegative.Conclusion: The mononuclear-macrophages can be induced to express the markers of lymphaticendothelial cells by FN, VEGF-C, TNF-α, LPS, IL-3, and IL-7.
