Objectives :To evaluate the expression of Matrix Metalloproteinase3(MMP-3) after artery injury and to explore the effect of MMP-3 on restenosis.Methods: 70 SD male rats were randomly divided into two groups,a group for the control group, and the other group for the experimental group. A rat model of vascular restenosis was established by balloon injury. The animals were harvested at 1,3,5,7,10,14,28 day after operation.the expression of MMP-3 was detected with HE staining and immunohistochemistry (IHC) staining. MMP-3 protein content was measured by western blot analysis at different time points in all groups.Results : 1 .HE staining The control group has no significant changes in intima, The experimental group has significantly artery wall thickening, mainly in intimal thickening. A large number of ECM and SMC in intima .2. Immunohistochemistry (IHC) staining MMP-3 in the balloon-injured rat artery intima and media of expression was strongly positive. The 1st day after Artery injury ,the expression began to increase, and reached a maximum at day 10,then declined. There are still some expression at day 28. The MMP-3 expression of rat thoracic aorta after balloon injury at different time points have significant differences (P <0 .01) between the control group and the experimental group. The control group at different time points do not exist significant difference(P> 0 .05), balloon injury group at different time points have significant difference(P <0 .01).Compared with the balloon injury group and control group, The expression of MMP-3 increased from the 1st day to the 28th day.Conclusions: The expression of MMP-3 increased after artery injury. MMP-3 may play important role in the early stage of restenosis through degrade collagen IV of basement membrance, which play an important role in migration and reproduction ofsmooth muscle cell in the early stage of restenosis.
