ObjectivesTo investigate the states of TEMT in HK-2 induced by high glucose in vitro,detect the protein expression of integrin-linked kinase(ILK)and matrix metalloproteinases-9(MMP-9),illustrate the correlation between ILK and MMP-9 in the process of TEMT.METHODSHuman proximal tubular epithelial cells(HK-2) were cultured in vitro and then divided into three group:normal glucose group(group N), high glucose group(group H),and high osmotic control group(group M). At 0h,24h,48h,72h,disposal cells were collected and total protein was extracted.The expression ofα-SMA,E-cadherin,ILK and MMP-9 were detected by Western blotting.RESULTS1.Compared with group N,the protein expression of a-SMA, E-cadherin had no significant changes in group M(P>0.05).2.Compared with group N,the protein expression of a-SMA,ILK and MMP-9 was increased(P<0.05) with a time-dependent mannar in group H,while the protein expression of E-cadherin was decreased (P<0.05) with a time-dependent mannar.3.The expression of ILK was inversely correlated with the expression of E-cadherin(r=-0.846,P<0.01),but which was directly correlated with expression ofα-SMA and MMP-9(r=0.950,P<0.01;r =0.982,P<0.01).CONCLUSION1.High glucose can induce TEMT in vitro.2.ILK participates and plays a positive control role in the process of TEMT.3.The expression of MMP-9 is up-regulated in high glucose-induced TEMT,which is highly related to the expression of ILK.
