Objective:To investigate whether human non-small cell lung cancer(NSCLC) tissue expresses granulocyte-colony stimulating factor receptor(G-CSFR) and whether G-CSFR plays an important role in the development, progression and prognosis of human NSCLC.To understand the role of granulocyte-colony stimulating factor(G-CSF) in the development and progression of NSCLC.Methods:Forty-eight specimens of tumor tissue and normal pulmonary tissue (taken 5cm apart from the tumor) were examined,which were taken from the NSCLC patients without neoadjuvant therapy.Immunohistochemistry and Western Blot techniques were employed to obtain G-CSFR protein expression,and reverse transcription polymerase chain reaction(RT-PCR) technique was used to catch G-CSFR mRNA expression.The relationship between expressions of G-CSFR and clinical or pathological factors was analyzed.Results:1.The immunohistochemical analysis showed that G-CSFR protein was found both in the cytoplasm and cytomembrane of the tumor tissue and normal tissue.The positive degree of G-CSFR protein in tumor tissue was higher than that in the normal tissue(P=0.019).The positive degree of G-CSFR protein was higher in weaklier differentiated tumor tissue. (P=0.008).There was no significant relation between the expressions of G-CSFR protein and the sex,age,minor size,histological classification and clinical stage(P=0.690,P=0.489,P=0.290,P=0.552,P=0.110)2.Western Blot revealed that the level of G-CSFR protein in tumor tissue was significantly higher than that in normal tissue(P=0.000).The level of G-CSFR protein was higher in the cases of poor differentiation and advanced clinical stage(P=0.008,P=0.000).There was no significant relation between the levels of G-CSFR protein and the sex,age,tumor size,histological classification(P=0.876,P=0.075,P=0.770,P=0.925 )3.RT-PCR revealed that the level of G-CSFR mRNA in tumor tissue was significantly higher than that in normal tissue(P=0.005).The level of G-CSFR protein was higher in poorer differentiated tumor tissue and the tumor tissue of advanced clinical stage(P=0.027,P=0.000).There was no significant relation between the levels of G-CSFR mRNA and the sex, age,tumor size,histological classification(P=0.980,P=0.162,P=0.460, P=0.611)4.The relationship between the G-CSFR mRNA and G-CSF protein was liner regression(P=0.000). Conclusions:1.G-CSFR protein was stained positively in the cytoplasm and cytomembrane.The level of G-CSFR protein and G-CSFR mRNA in tumor tissue were higher than those in the normal tissue of patients with NSCLC.2.The level of G-CSFR and G-CSFR mRNA were higher in the cases of advanced stage and poor differentiation,which indicated that G-CSFR may play an important role in the development and evolution of NSCLS and it might be related to poor prognosis of NSCLC.
