OBJECTIVE:Analysis IVF-ET/ICSI-ET clinical outcome of the patients with diminished ovarian reserve.Research the relative amount of copy number of granular cell mitochondria DNA(mtDNA) and deletion rate of 4977bp for DOR patients in order to primarily study structural integrity of granular cell mitochondria DNA(mtDNA).MATERIALS AND METHODS:Respective 50 patients of DOR and NOR groups were selected from Reproduction Centre of Xiangya Hospital from January to October 2008,Compare the difference of clinical outcome within the two groups.Granular cells in liquor folliculi of these patients were collected at ovum pick-up day,Then,DNA was extracted from the granular cells by the conventional phenol:chloroform. The mtDNA 4977bp deletion of granular cell were detected by PCR while relative quantity of granular cells mtDNA copies were detected by real-time PCR.Compare the difference of mtDNA4977bp deletion of granular cells and mtDNA relative quantity copies of the two groups to verify whether there is any statistical significance.RESULTS:1.The oocytes quantity(5.86±3.50 vs.18.64±7.39),zygote quantity (69.28%vs.75.43%),High-quality embryos(2.72±1.93 vs.7.48±4.67) and clinical pregnancy rate(8%vs.54%) of DOR group were obviously lower than NOR Group. 2.No 4977bp deletion of ovary granular cell mitochondria DNA of 100 patients in the DOR and NOR groups was detected.3.The multiples of copy number to the contrast sample are 0.32496~1.16384 in DOR group and 0.31969~1.26171 in NOR group, respectively,after t-text,there is no significance of relative quantity difference of granular cell mitochondria DNA in DOR and NOR groups,(t=1.146;P=0.255>0.05).CONCLUSION:1,Poor result of clinical outcome in vitro fertilization-embryo transfer treatment for DOR patients.2.The structure of DOR patients' granular cells mtDNA is basic integrity,maybe unrelated with the above conclusion.3.Granular cell may be used as donor cells for DOR patients plasma autologous transplants mitochondorial.
