Effects Of DRD₁,DRD₂ Polymorphisms On Antipsychotics Responses In Schizophrenic Patients

Aims:The varible responses to antipsychotic drugs in the treatment of schizophrenia still present obstacles to tailored medicine.The present study aimed to clarify the distribution feature of DRD₁ G685A and DRD₂ G-607T G-1409T polymorphisms in Chinese Han population and investigate the association between DRD₁ G685A and DRD₂ G-607T G-1409T polymorphisms and risperidone and quetiapine response in patients.Methods:The clinical data of the patients were investigated with the inventories.The severity of symptoms and the responses to risperidone and quetiapine were rated by Brief Psychiatric Rating Scale(BPRS). Pyrosequencing and asPCR were applied for genotyping.T-test,Chi-square(x²) test,Fisher exact test and Wilcoxon rank sum test were applied for statistical analysis.Results:(1) The A allelic ofDRD₁ G685A in Chinese was not found.(2) The two SNPs of DRD₂ gene were in Hardy-Weinberg equilibrium.(3) The T allelic frequency of G-607T in patients and health control were 45.8%and 44.0%,respectively.No significant difference was observed between two groups. After risperidone treatment,the reduction rate of BPRS score of patients with GT+TT genotypes was higher than that with GG genotype(P＜0.05).After quetiapine treatment,no significant difference was observed between two groups.(4) The T allelic frequency of G-1409T in patients and health control were 49.5%and 44.3%,respectively,No significant difference was observed between two groups.After risperidone treatment,the reduction rate of BPRS score of patients with GT+TT genotypes was higher than that with GG genotype (P＜0.01).After quetiapine treatment,no significant difference was observed between two groups.Conclusions:There was no polymorphism of DRD₁ G685A in Chinese. These data suggested that DRD₂ G-607T and G-1409T polymorphisms might influence the response to risperidone in Chinese Han patients.Impaired therapeutic efficancy of risperidone might be found in patients carried mutation allele.There might be no relationship between polymorphisms and quetiapine response.
