Obejectives Lung adenocarcinoma is one of the most common types of maliganat tumors in the world,threatening people's health with increasing impact.While we have not developed effective treatment to it. The theory of cancer stem cells brings new promise for us to understand and treat tumors.Scientists have continuously sorted cancer stem cells for many kinds of tumors according to the protein markers on cell surface. However,till now phenotype of cancer stem cells for human lung adenocarcinoma have not been identified.It is the premise of sorting, identifying and studying cancer stem cells efficiently to select appropriate combination of surface markers.This study provides following steps of sorting cancer stem cells with evidence and offer reference to indentifying surface protein markers for cancer stem cells for human lung adenocarcinoma.Methods In this study we use two kinds of culturing methods to generate both adhesive tumor cells and floating aggregates,then compare expression of CD133,CD34 and CD44 between these two kinds of cells. Because it is known that there are high percentage of cancer stem cells in floating aggregates.Difference of expression of these 3 kinds of surface protein markers between 2 kinds of cells would indicate whether CD133, CD34 and CD44 could be used as appropriate combination of biomarkers for cancer stem cells for human lung adenocarcinoma.Results When culturing cells,we have found that floating aggregates grow more slowly,keep activity for longer period than adhesive cells. However we successfully generate floating aggregates in less than 20% specimens,while 68%for adhesive cells.In the immunofluorescence test, floating aggregates express much higher level of CD133,CD34 and CD44 than adhesive cells.As a result,we consider that CD133,CD34 and CD44 can be used as appropriate combination of surface markers for cancer stem cells for human lung adenocarcinoma.Conclusion CD133,CD34 and CD44 can be used as appropriate combination of surface markers of cancer stem cells for human lung adenocarcinoma.
