The Expression And Significance Of WIF-1 In The Cartilage In Patients With Osteoarthritis

Objective:This study is designed to measure the contents of WIF-1 in the cartilage with osteoarthritis(OA),in order to demonstrate the relationship of WIF-1 and osteoarthritis(OA),and study the effect on OA pathogenesis and the clinical significance,as to give a theoretical evidence for prevention and treatment for OA.Methods:Human cartilages were obtained at arthroplasty from 30 patients with OA.Corresponding articular cartilages were obtained at arthroplasty or amputation for trauma,tumour or infection from 10 patients with no history of arthritis.Then collect 48 cartilage specimens from OA and normal joint,all the specimens were fixed,decalcified, embedded in paraffin and cut into slices.Each slices sample was stained with HE and Safranin O,on the basis of improved Mankin pathological score to divide into four groups,normal group 10 specimens(group A), mild impaired group 9 specimens(group B),medium impaired group 14 specimens(group C),severe impaired group 15 specimens(group D). Meanwhile,the slices were treated by immunohistochemical reaction of WIF-1 and dealed with micrograph analyze instrument(MIAS system). Each slice was randomly mensured 4 high fields of eyeshot,and made semi-quantitative analysis of average optical density(AOD).The data was analyzed with Spearman rank correlative analysis and analysis of variance,performed in SPSS Stat software.P values less than 0.05 were considered statisticaly significant. Results:1.WIF-1 expression in both normal and OA human cartilage in all layer,and the distribution is around the Chondrocytes scattered.2.The AOD of WIF-1 in the group A is 0.2603±0.0396,group B 0.2412±0.0341,group C 0.1881±0.0444 and group D 0.1638±0.0385 There are significant differences between osteoarthritis group and nomal group(p＜0.05).There are no significant differences between group A and group B(P＞0.05),While there are significant differences between group A and group C&D(p＜0.01).there are significant differences between group B and group C&D(p＜0.05),There are no significant differences between group C and group D(P＞0.05).According to Spearman rank correlation test,the AOD of WIF-1 have direct correlation with the improved Mankin pathological score(r=-0.896,p＜0.05).Conclusion:1.The first time discovered that WIF-1 expression in normal human cartilage and OA cartilage.2.WIF-1 low expression is one of important pathogenesis in OA.3.With the expression of WIF-1 decreases,the cartalege of OA impaired increases by degrees.
