Objectives:To explore the expression of vascular endothelial growth factor(VEGF) and Kinase domain receptor(KDR)in human prostate cancer(PCa),and to explore their relationship with tumor gleason score,pathological grading and its clinical staging in PCa.Methods:To order to understand the differences of VEGF,KDR expressions among the normal human prostate,benign prostatic hyperplasia and prostate cancer and to seek out the relationships between the various pathological ratings of prostate cancer and VEGF,KDR as well as the relationships between clinical phases and Gleason scoring,researchers stained 10 cases of normal specimens and 15 cases of benign prostatic hyperplasia specimens and 40 cases of prostate cancer specimens.The expression of VEGF,KDR was dectected by means of immunohistochemistry.The principle of immunohistochemical staining:There are some different immunological reactions between marked antibodies and antigens in histiocytes.Then,tumor markers on marked antibodies react to developers.Last,we can definitely detected antigens quantity and cytological location by observing chromogenic objects with microscope.Results:1.The positive expressions of VEGF,KDR in prostatic tissues are stained from yellow to brown in cytoplasm.Only two positive expressions of VEGF and only one positive expressions of KDR in normal prostate and the color is lighter yellow,the positive rate of VEGF and KDR in benign prostatic hyperplasia is 33.3%and 26.6%respectively,but the positive rate in prostate cancer reach to 87.5%and 90.0%, whose color in cytoplasm is deeper.The positive rates of VEGF and KDR in PCa was higher than those in BPH and NP(P<0.05).2.The positive rates of VEGF and KDR in high Gleason score group were higher than those in low Gleason score group. 3.All of the negativeVEGF and KDR expressions are distributed in well differented tumors,the highly positive rate of VEGF and KDR in poorly differented tumors are 85.7%and 78.5%;62.5%and 75.0%negative expression in A staging; 100%positive expression in D staging,Moreover,75.0%and 66.7%highly positive expression in D staging.The postive rate of VEGF and KDR correspond to the increase of tumor's pathological grading and its clinical staging and furthermore shows deeper color in cytoplasm.4.Using Spearman rank correlation analysis,there was a positive correlation between the expression of VEGF and that of KDR(r_s'=0.5995,P<0.01).Conclusions:1.The expression of VEGF and KDR in prostate cancer is obviously higher than in normal human prostate and in benign prostatic hyperplasia.2.The positive rates of VEGF and KDR in high Gleason score group were higher than those in low Gleason score group.3.The expression of VEGF and KDR in prostate cancer presents positive correlation with tumor's grading and clinical staging.The expression presents close relation to the carcinogenesis,progression and metastasis of prostate cancer.
